荧光定量RT-PCR检测性成熟前与性成熟山羊卵巢α/β-ER mRNA表达的比较

摘  要:为分析性成熟前与性成熟波尔山羊和沂蒙黑山羊卵巢内α、β雌激素受体(α/β-ER)表达的差异规律,根据已发表波尔山羊α/β-ER基因序列,设计不同引物,对PCR反应条件进行优化,建立稳定检测2种山羊卵巢雌激素受体mRNA含量的荧光定量RT-PCR方法;并对性成熟前后2种山羊卵巢组织中α/β-ER mRNA进行了检测比较。结果显示,性成熟前2种山羊卵巢α-ER和β-ER mRNA表达含量较性成熟山羊低;而且这2种山羊无论性成熟前还是性成熟后,α雌激素受体表达均显著低于β雌激素受体。To compared the mRNA expression of α/β-estrogen receptors(ERα.ERβ) in ovaries ob- tained trom Boer and Yimeng black goats in prepuberty and puberty, 6 couples of α/β-ER gene primers were designed and synthesized; the conditions of reaction such as annealing temperature and time, extending temperature and time, cycle times were optimized with gradient PCR or PCR assays;the mRNA expression of ERα, ERβ in ovary follioles were detected with above-optioned fluorescence quantitative RT-PCT assays. The results indicate that the mRNA expressions of ERα/ERβ in prepubertal ovaries of Boer goats and Yimeng black goats are less than those in pu- bertal ones, and the mRNA expression of ERa in ovaries of both goats, whether in prepuberty or in puberty, are less that of ER;. These results will be helpful to clarify the mechanism on different developmental competence of goats follicle oocytes in prepuberty and in puberty.
