摘  要:目的:分析人子宫珠蛋白与猫主要过敏原Fel d 1之间关系。方法 :运用ClustalW 1.83和MEGA5.1 Beta3软件比较人分泌球蛋白家族序列及猫过敏原Fel d 1两个亚基P30438和P30440之间的进化树形图;采用在线软件平台NetMHCII 2.2比较分析P30438与子宫珠蛋白典型序列P11684之间抗原性差异;利用Swiss-Model程序模拟并比较P11684、P30438和P30440的三级结构。结果 :分泌球蛋白家族充斥着大量冗余数据,经逐步聚类收缩至13个代表性序列进一步分析发现,整个分泌球蛋白家族划分成两个大家族三个亚家族,而子宫珠蛋白典型序列与猫主要过敏原Fel d 1的一个亚基P30438之亲缘关系最近,两者均具有与HLA高亲和力结合位点,而且三级结构相似。结论:子宫珠蛋白具有潜在的过敏原性。Objective To explore the relationship between human uteroglobin and cat allergen Fel d 1.Methods Bioinformatic software such as ClustalW 1.83 and MEGA5.1 Beta3 were employed,for sequence alignment and clustering,on the sequences of secretoglobins and the two subunits(P30438 and P30440) of Fel d 1 retrieved from NCBI and SwissProt,supplemented with manual subtraction of accession items.The antigenicity of P30438 and P11684,a typical sequence of uteroglobin were compared by online bioinformatic software NetMHCII 2.2.The 3D structures of P30438,P30440 and P11684 were modeled and deciphered in parallel.Results Secretoglobin family harbored a large amount of redundant sequences.These sequences were first classified into 7 subgroups and were then by progressive clustering reduced to 13 representative non-redundant sequences.P11684 was closely related to the subunit P30438 of Fel d 1.P11684 and P30438 both displayed several high binding affinities with HLA molecules.The 3D structure of P11684 was similar with those of P30438 and P30440.Conclusion Uteroglobin may have the potential allergenicity,as cat allergen Fel d 1 does.
