摘 要:目的:建立简便的耐受性小鼠骨髓源性树突状细胞(dendriticcells,DCs)的诱导培养,并探讨耐受性DCs的生物学功能。方法:直接冲出小鼠骨髓源细胞,用IL-4和GM-CSF诱导分化,对终细胞进行形态的观察、表型的鉴定、和功能检测。结果:诱导培养7d的DCs低表达表面共刺激分子为非成熟状态,非成熟DCs也可刺激淋巴细胞增殖并且分泌的IL-10显著升高。结论:这种获得耐受性小鼠骨髓源性DCs的方法相对简便,耐受性DCs可刺激淋巴细胞增殖且高分泌IL-10。Objective To explore a relatively simple method of inductive cultivation of tolerant bone marrowderived dendritic cells(DCs) of mice and to investigate the biological function of tolerant DCs.Methods Bone marrow-derived DCs were induced by GM-CSF and IL-4 from the culture of mice bone marrow cells in vitro.The morphology,phenotype,and function of DCs were observed.Results DCs with low expression of surface costimulate molecular in day 7 were in immature state,these DCs could stimulate the proliferation of lymphocytes and the secretion of IL-10.Conclusion The method used in this study was a simple method to harvest tolerant bone marrow-derived DCs,which could stimulate the proliferation of lymphocytes and the secretion of IL-10.
