摘 要:基因表达谱芯片是对一样本同时进行上千基因表达检测的技术。通过对不同种已知免疫毒性物质的表达谱变化研究,逐步建立了以发现已知或未知免疫毒性、预测外源物质暴露后生物系统的免疫毒性反应、对免疫毒性物质分类为研究目的免疫毒性研究方向。与此同时,全球数据库及基因组学研究的试验标准化、新技术如树突状细胞的体外培养等,对研究基因表达谱芯片在免疫毒性研究开拓了新方法和思路。本文将从以不同研究方向下的具体应用入手简述免疫毒性基因表达谱芯片的应用。Gene expression microarray is the technique to detect gene expression of thousands of genes simultaneously in one sample.Through analyzing gene expression profiles of immunomodulating agents,we gradually study immunotoxicity dividing it into three parts by purposes as follows: detecting known and novel effects of a wide range of immunomodulating agents,predicting consequences of exposing biological systems to toxic agents and classifying compounds.Besides,those better databases and guidelines have been settled globally,and the new technologies such as the isolation and culture of dendritic cells from blood combining the gene expression profiles have been developed.They provide new method and insight in preclinical and clinical immunotoxicity study in known and novel immunotoxicity substances.Here we introduced the applications of gene expression microarray in immunotoxicity under the different directions of the specific application.
