补肾健脾方对去卵巢大鼠骨质疏松症细胞因子水平的影响  被引量:10

摘  要:目的:通过复制去卵巢大鼠所致的骨质疏松模型,研究补肾健脾方对去卵巢大鼠骨质疏松症细胞因子水平的影响。方法:选取6月龄雌性SD大鼠60只,按体质量随机分为4组,即假手术组、去卵巢组、阳性对照组及补肾健脾方组,每组15只。除假手术组外,其余各组按文献方法切除大鼠双侧卵巢,造成雌激素缺乏而致的实验性骨质疏松症,假手术组手术过程与卵巢切除组相同,但不切除卵巢而只切除小块脂肪组织。术后1周开始灌胃给药,灌胃12周后所有大鼠股动脉取血,收集血清,采用ELISA法检测血清IL-6、IL-4、TNF-a、IGF-1水平。结果:去卵巢组大鼠IL-6和TNF-a水平升高,IL-4和IGF-1水平降低,差异有统计学意义(P<0.05)。与去卵巢组比较,补肾健脾方组能降低IL-6和TNF-a水平,提高IL-4和IGF-1水平,差异有统计学意义(P<0.05)。结论:补肾健脾方通过降低IL-6和TNF-a水平,提高IL-4和IGF-1水平,防治去卵巢所致大鼠的骨质疏松。Objective: To study the effect of Bu Shen Jian Pi decoction on cytokine through the osteoporosis models of the rats. Methods: Sixty 6-month-old female Sprague-Dawley rats were used and divided into following groups by body weight randomly: sham control group, ovx control group, ovx with nylestriol, ovx with Bu Shen Jian Pi decoction. Bilateral ovariectomies were conducted under pentobarbital anesthesia on the OVX, Bu Shen Jian Pi decoction and NE groups. A sham operation, during which the ovaries were touched with forceps, was performed on the sham group. The treatment began 1 week after the surgery and lasted for 12 weeks. 12 weeks later, serum was separated. The level of IL-6, IL-4, TNF-a and IGF-1 were measured via the method of ELISA. Results : Ovx control group showed the lower level of IL-4 and IGF-1, IL-6 and TNF-a increased levels, there was a significant difference (P 〈 0.05). Nylestriol and Bu Shen Jian Pi decoction groups reduced IL-6 and TNF-a, increased IL-4 and IGF-1, compared with ovx control group have significant difference (P 〈 0. 05). Conclusion: Bu Shen Jian Pi decoction is effective for treating osteoporosis of ovariectomized rats. It can lower IL-6 and TNF-a levels and raise IL-4 and IGF-1 levels.
