益气健脾方药对脾虚大鼠脑内JAK1、STAT1、SOCS1水平变化的影响

摘　　要：目的:观察益气健脾方药对脾虚大鼠脑内Janus激酶1(JAK1)、信号转导和转录激活因子1(STAT1)、细胞因子信号抑制分子1(SOCS1)水平变化的影响。方法:将清洁级雄性Wistar大鼠80只,随机分为正常组、模型组、治疗1组和治疗2组,除正常组外其余3组造脾虚模型。治疗1组合治疗2组分别予四君子汤和玉屏风散灌胃治疗。6周后灌注取脑,石蜡包埋,切片处理后采用免疫组化法检测脑内JAK1、STAT1和SOCS1的水平变化。结果:脾虚组大鼠脑内海马CA1区及下丘脑腹侧核的JAK1和STAT1的水平明显升高,SOCS1水平明显降低,与正常组比较差异显著(P<0.05);治疗1组大鼠在海马CA1区及下丘脑腹侧核的JAK1和STAT1水平较低,SOCS1的表达水平较高,与模型组比较差异显著(P<0.05),而与正常组比较无显著差异;治疗2组大鼠在海马CA1区和下丘脑腹侧核JAK1、STAT1和SOCS1的水平与正常组和治疗1组比较无显著差异。结论:益气健脾方药四君子汤、玉屏风散可通过影响JAK-STAT信号通路起到改善机体免疫功能,提高免疫能力的作用。Objective：To obsearve the influence of Four Gentlemen Decoction and Jade-Screen Powder on the expressions of Janus kinase 1（JAK1）,signal transducer and activator of transcription 1（STAT1） and suppressor of cytokine signaling 1（SOCS1） in the brain of spleen deficiency（SD） model rats.Methods：The experiment used Wistar rats as the subjects,and they were divided into four groups： normal group,model group,treatment group 1（model rats with Four Gentlemen Decoction）,treatment group 2（model rats with Jade-Screen Powder）.This experiment aimed to further unfold the influence of spleen deficiency on body defense function,using immunohistochemical study to detect the amounts of JAK1,STAT1 and SOCS1 in the brain of SD rats.Results：Compared with normal group,the expressions of JAK1 and STAT1 were increased in both hippocampal CA1 area and ventral hypothalamus of model group,while the expression of SOCS1 was decreased（P〈0.05）.Compared with model group,the expressions of JAK1 and STAT1 were decreased in both hippocampal CA1 area and ventral hypothalamus of treatment group 1,while the expression of SOCS1 was increased（P〈0.05）.There was no obvious variance among normal group,treatment group 1 and treatment group 2.Conclusion：Four Gentlemen Decoction and Jade-Screen Powder can improve human defense function through influencing the genetic expression of JAK-STAT signal pathway so as to strengthen human defense capability.
