摘  要:目的采用毛细管电泳-安培检测(CE-AD)法测定白头翁及其多糖水解物中半乳糖、葡萄糖、鼠李糖、阿拉伯糖、木糖、甘露醇、蔗糖等组分。方法采用热水浸提、乙醇沉淀的方法,提取、分离白头翁中甘露醇及糖类组分。CE-AD采用25μm×80 cm石英毛细管,工作电极300μm铜圆盘电极,检测电位0.70 V[vs.饱和甘汞电极(SCE)]。结果在运行缓冲液为0.08 mol/L NaOH,分离电压为16 kV,电动进样8 s的检测条件下,待测组分可达到基线分离与灵敏检测,待测组分浓度在5.0×10-6~5.0×10-4mol/L范围内与CE-AD信号呈良好线性关系。白头翁提取液中含有甘露醇、蔗糖、半乳糖、葡萄糖、鼠李糖、阿拉伯糖、木糖,摩尔比在1.0~55.6范围内;白头翁多糖水解物中含有甘露醇、半乳糖、葡萄糖、鼠李糖、阿拉伯糖、木糖,摩尔比在0.2~18.8范围内。结论该方法准确、重现性较好,可用于白头翁药材中糖类组分的分析检测。AIM To adopt the capillary electrophoresis-amperometric detection(CE-AD) for determining galactose,glucose,rhamnose,arabinose,xylose,mannitol and sucrose in Pulsatillae Radix and its polysaccharide hydrolysate.METHODS Mannitol and saccharides in Pulsatillae Radix were extracted and purified with hot-water-extracting and alcohol-precipitating operation.CE-AD was performed with a 25 μm × 80 cm quartz capillary and a 300 μm copper disk electrode as working electrode with +0.70 V [versus the saturated calomel electrode(SCE)].RESULTS With a running buffer of 0.08 mol/L NaOH,separation voltage of 16 kV,electrokinetic injection of 8 s,the components could reach baseline separation and sensitive detection.The concentrations of components were linear correlation with the corresponding CE-AD signals in the range of 5.0×10-6-5.0×10-4 mol/L.Mannitol,sucrose,galactose,glucose,rhamnose,arabinose and xylose were found in the extract of Pulsatillae Radix with a molar ratio of 1.0-55.6.The hydrolysate of polysaccharide was constituted of mannitol,galactose,glucose,rhamnose,arabinose and xylose with a molar ratio of 0.2-18.8.CONCLUSION The proposed CE-AD method could be applied to the determination of saccharides in Pulsatillae Radix with satisfactory accuracy and reproducibility.
