摘 要:猪瘟是由猪瘟病毒引起的猪的高度致死性、接触性传染病。为了解CSFV毒株对猪体免疫系统的影响,本研究使用中等致病力CSFV毒株人工感染试验猪,定期采集猪外周血,提取总RNA,运用qRT-PCR技术对免疫相关因子的mRNA转录水平进行分析。试验结果表明,在临床症状早期和在临床症状明显期,11种细胞因子的转录水平均有不同程度上升(RQ>1);在濒死期,除AMCF、IL-1α、IL-1β、IL-6转录升高外,其余细胞因子转录水平下降显著(RQ<0.1)。该试验结果证实,CSFV感染可以导致PBMC细胞分泌的细胞因子mRNA转录水平增高,从而影响机体的细胞免疫应答和体液免疫应答功能,这可能是CSFV对猪免疫功能影响的分子机制之一。Classical swine fever (CSF) is a high morbid and contagious disease caused by the agent of classical swine fever virus (CSFV). Host defense mechanisms for the severe outcome of different types of CSFV infections have not been clearly known. To understand these mechanisms , the transcriptional levels of IL-1, IL-2, IL-4, IL-6, IL-8, IFN-α, IFN-β, IFN-γ, MxA, PKR, OAS, AMCF-1, AMCF-2 and TNF of PBMC derived from piglets experimentally inoculated with CSFV at 40 days old were detected by quantitative RT-PCR(qRT-PGR). The results showed that all cytokines mRNA levels detected in this study were notably increased from 1 to 21dpi (RQ〉1) and decreased significantly in agonal stage except for AMCF, IL- 1α, IL-1β, and IL6. Data suggest that the observed gene expression profile might explain immunological and pathological changes associated with CSFV infection.
