摘  要:目的:观察白藜芦醇对肾性高血压大鼠脑动脉瘤形成的抑制作用及其对NF-κB活性的影响。方法:制作大鼠脑动脉瘤模型,随机分为对照组、模型组和治疗组,光镜及电镜观察组织学变化,应用免疫组化及EMSA检测NF-κB的动态变化。结果:组织学观察中对照组没有动脉瘤产生,模型组动脉瘤发生率约12%,治疗组大鼠给予白藜芦醇30mg/kg,仅发现部分动脉呈瘤前病变,无明显的动脉瘤产生。NF-κB在各组脑动脉壁中均有表达,对照组NF-κB活性微弱;模型组随着大鼠血压的增加其NF-κB活性迅速增加,术后1月时其活性增至高峰,并持续至术后2月,以后逐渐下降,术后4月降至正常水平;治疗组NF-κB活性被明显抑制,随着大鼠血压的增加,NF-κB活性仅有轻度增加,无明显的活性高峰出现。结论:NF-κB的高表达在脑动脉瘤形成的发病机制中起重要作用,白藜芦醇抑制NF-κB的高表达可能是防止动脉瘤的发生的机制之一。Objective: To investigate the mechanism of Resveratrol on preventing cerebral aneurysm induced by renal hypertension in rats and the effect on the activity of NF-κB.Methods: A rat model of cerebral aneurysm was established,the rats were divided into three groups,control group,model group and treated group.Histologic changes of cerebral arteries were observed by light and electron microscopy,The dynamic change of NF-κB were examined by EMSA and Immunohistochemistry.Results: No aneurysm was found in control group,there was little NF-κB activity in cerebral artery;12% rats of model group had cerebral aneurysms,with the development of hypertension,The NF-κB activity in cerebral artery increased apparently,peaked at one month after operation,lasted two months,returned to baseline at four month postoperation.Only preaneurysmal changes were found in treated group,The NF-κB activity in cerebral artery was inhibited obviously,with the development of hypertension,The NF-κB activity increased slightly,there was no activity peak.Conclusion: High expression of NF-κB plays an important role in the pathogenesis of cerebral aneurysms.Resveratrol can inhibit the expression of NF-κB.Maybe it's one of the mechanisms that resveratrol prevents hypertensive aneurysm formation.
