摘  要:目的:探讨不同剂量槲皮素对非酒精性脂肪肝(Non-alcoholic fatty liver disease,NAFLD)大鼠血清白介素18(IL-18)和白介素10(IL-10)含量的影响。方法:140只SD大鼠随机分为正常对照组、模型组、槲皮素高剂量治疗组(300 mg/kg)、槲皮素低剂量治疗组(75 mg/kg)。采用高脂饲料构建非酒精性脂肪肝大鼠动物模型,并用槲皮素进行干预治疗,分别于给药后4周、8周、12周末,采用ELISA法检测大鼠血清中IL-18、IL-10的含量。并进一步检测给药后8周末大鼠肝指数、甘油三酯(TG)和空腹血糖(FBS)情况。结果:模型组与对照组比较血清IL-18、IL-10从第4周开始即有明显变化(P<0.05或P<0.01),而且各时间段之间比较有统计学差异(P<0.05);槲皮素300 mg/kg、75 mg/kg与模型组比较,血清IL-18从第8周开始明显降低(P<0.01),血清IL-10从第4周开始显著升高(P<0.05或P<0.01);槲皮素不同剂量组之间比较:血清IL-18从第8周开始高剂量组降低明显,与低剂量组之间有统计学差异(P<0.01),但血清IL-10水平不同剂量组之间均无统计学意义(P>0.05)。槲皮素灌胃治疗8周后,各治疗组与模型组比较:肝指数、FBS均显著下降(P<0.01或P<0.05),TG水平下降仅高剂量组有统计学意义(P<0.01);不同剂量治疗组之间比较:高剂量槲皮素组比低剂量组FBS、TG水平降低有统计学意义(P<0.01),但肝指数变化无差异(P>0.05)。结论:血清IL-18和IL-10水平与NAFLD密切相关,提示促炎和抗炎细胞因子在NAFLD发病中可能起重要作用。槲皮素可使血清IL-18水平降低、IL-10升高,可能通过缓解炎症因子失衡,从而对NAFLD起到一定的治疗作用。Objective: To study the effects of different dosages of quercetin on serum IL-18 and IL-10 of nonalcoholic fatty liver disease(NAFLD) in rats.Methods: 140 SD rats were randomly divided into the control group,model group,high dosage of quercetin(300 mg/(kg·d)) group and low dosage of quercetin(75 mg/(kg·d)) group.To construct the animal model of NAFLD in rats with high fat diet,and different dosages of quercetin were administrated.The serum IL-18 and IL-10 in the rats were determined by ELISA at 4 weeks,8 weeks and 12 weeks after the administration of quercetin.Furthermore,liver index,triglyeride(TG) and fasting blood sugar(FBS) were detected after 8 weeks after the administration.Results: Compared with the control group: The level of serum IL-18 and IL-10 in model group were significantly changed from 4 weeks after the administration of quercerin(P0.05 or P0.01),and there was statistical difference among different time(P0.05).Compared with the model group: The level of serum IL-18 in the different dosages(300 mg/(kg·d) and 75 mg/(kg·d)) of quercetin groups was dramatically decreased from 8 weeks after the administration,and IL-10 was increased from 4 weeks after that(P0.05 or P0.01).The level of IL-18 in the high dose group was lower than that in the low dose one from 8 weeks after the administration(P0.01),but the level of IL-10 was no statistical difference between different dosages groups(P0.05).At the 8 weeks after the administration of quercetin: The liver index and FBS in different dosages groups were obviously decreased compared with the model group(P0.05 or P0.01),however,TG was significantly decreased only in the high dosage group(P0.01).The FBS and TG in high dose group were obviously lower than that in the low dose one(P0.01),but there was no dramatically difference in the liver index between different dosages groups(P0.05).Conclusion: The amount of IL-18 and IL-10 in serum was closely related with NAFLD.The c
