摘 要:目的:观察结肠康对恶唑酮(Oxazolone,OXZ)诱导小鼠溃疡性结肠炎(Ulcerative colitis,UC)炎症反应的抑制作用。方法:小鼠颈背部3%恶唑酮致敏2天,第6天用1.5%恶唑酮灌肠,灌肠当日开始灌胃不同剂量结肠康。灌胃3天后,断头处死小鼠,酶联免疫吸附法(ELISA)测定结肠组织匀浆液中炎性因子IL-4、TNF-α、IL-1β和IL-10浓度。柳氮磺胺吡啶作为阳性对照药。结果:与空白对照组相比模型组IL-4浓度降低64.64%,TNF-α、IL-1β和IL-10浓度增加121.03%、73.04%和213.13%。结肠康6.4、12.8和25.6g/kg组能显著增高IL-4的浓度,降低TNF-α、IL-1β和IL-10浓度,实验结果提示结肠康0.32g/ml、0.64 g/ml和1.28g/ml呈剂量依赖性改善结肠炎症反应细胞因子浓度,结肠康1.28 g/ml剂量组治疗效果最好与阳性对照药SASP无显著差别。结论:结肠康能抑制OXZ诱导小鼠溃疡性结肠炎炎症反应,对UC具有一定得治疗作用。Objective: To observe the inhibitory effects of Jiechangkang on inflamm-atory response of colon in oxazolone-induced ulcerative colitis(UC) mice.Methods:The allergy was produced by dropping 3% oxazolone on mice's skin and subsequently being intragastric administration 1.5% oxazolone on 6th day after sensit-ization.Simultaneously,the mice were begun to orally administrate three doses of Jiechangkang,and decapitated on the day 3 after administration.Enzyme-linked immunoadsordent assay(ELISA) was used to detect the levels of IL-1,IL-4,IL-10 and TNF-α of colon tissue.Willow nitrogen sulfonamides pyridine(SASP) was as reference drug.Results: Compared with the control group,IL-4 concentration decreased 64.64%,TNF-α,IL-1β,and IL-10 concentration increased 121.03%,73.04 % and 213.13%,respectively in colon tissue of UC mice induced by oxazolone.However,the decreased IL-4 and the increased IL-10,TNF-α and IL-1β were partly reversed by three doses of 6.4,12.8 and 25.6 g/kg Jiechangkang.The experimental results suggest Jiechangkang 0.32g/ml 0.64 g/ml and 1.28 g/ml dose dependent concentrations improve colonic inflammation reaction,1.28 g/ml dose group Jiechangkang therapeutic effect preferably and there was no significant difference in the positive control drug SASP.Conclusion: The present study gets a result that Jiechangkang has a certain effects on inflammatory response,which suggests that Jiechangkang may have benefit effect in treatment of UC.
