摘 要:目的检测低剂量饮酒大鼠肝组织DNA中氧化标志物8-羟基脱氧鸟苷(8-oxo-dGsn)水平,评价低剂量饮酒对肝脏组织氧化应激的影响。方法 4月龄的健康雄性SD大鼠48只,随机分为对照组和饮酒组,饮酒组每天灌胃56度二锅头酒每天每公斤低于2g(<2g/kg),对照组每天灌胃同等体积的生理盐水。在灌胃4、8、12周时,用放射性核素稀释高效液相-串联质谱(ID-LC-MS/MS)方法检测大鼠肝组织DNA中氧化标志物8-羟基脱氧鸟苷(8-oxo-dGsn)含量;用分光光度法测定抗超氧阴离子(O2-)能力和谷胱甘肽过氧化物酶(GSH-PX)活力。结果低剂量饮酒4、8、12周后的肝组织DNA中8-oxo-dGsn含量均较同时间点对照组低,差异有统计学意义(P<0.05);低剂量饮酒4、8、12周后的肝组织中抗O2-能力与GSH-PX活性均增强,与对照组相比差异有统计学意义(P<0.05)。结论低剂量饮酒可抑制大鼠肝组织DNA中的氧化水平,可能与肝脏中抗氧化酶活性的增高有关。Objective To study the level of 8 - dihydro - 2' - deoxyguanosine (8 - oxo - dGsn) , a biomarker of DNA oxidation, in the livers of rats after mild alcohol consumption. Methods Totally 48 male Sprague - Dawley (SD) rats at four months of age were allocated randomly in to two groups: the normal control group and the alcohol consumption group. The rats in the alcohol group were given 56° alcohol by intragastric administration in a total dose of 2g/( kg · d) while the the rats in the control group were given the same volume of physiological saline (0.9% , NaC1). At the end of 4, 8, 12 weeks of treatment, we determined the level of 8 -oxo- dGsn, as a bio- marker of DNA oxidation, in livers of rats by ID - LC - MS/MS, and meanwhile, detected the ability of agonistie - superoxide anion and the active of glutathion peroxidase ( GSH - PX) by spectrophotometric method. Results The level of 8 - oxo - dGsn in the liver tissues of alcohol group was lower than that of the control group in 4, 8 and 12weeks (P 〈 0.05 ). At the same time, the ability of agonistic - superoxide anion( O2- ) and the active of glutathion peroxidase ( GSH - PX) in the liver tissues of alcohol group were higher than that of control group (P 〈 0. 05 ). Conclusion These results suggest that a low dosage of alcohol consumption for rats would suppress the DNA oxidation in livers, which may be related to the active of the antioxidative enzyme raised up after drinking slightly.
