摘  要:目的:研究弓形虫感染人包皮成纤维细胞(HFFs)的microRNAs(miRNAs)差异表达。方法:基因芯片分析弓形虫感染的HFFs的miRNAs差异表达,并用实时荧光定量RT-PCR法和Northern blot法验证miRNAs差异表达。结果:弓形虫感染HFFs的46种miRNAs表达量有1倍以上上调,37种miRNAs表达量有1倍以上下调。其中4种差异表达的miRNAs的实时荧光定量RT-PCR法和Northern blot法验证结果与基因芯片分析结果一致。结论:弓形虫感染可导致宿主细胞的miRNAs差异表达,差异表达的miRNAs可能在弓形虫和宿主细胞的相互作用中起调控作用。Objective: To investigate microRNAs differential expression of human foreskin fibroblasts (HFFs) during Toxoplasma gondii infection. Methods: The microRNAs differential expression of HFFs during Toxoplasma gondii infection was analyzed by microarray, and further validated by quantitative realtime RT-PCR and Northern blot. Results: The result demonstrated that the level of 46 mature miRNAs upregulated more than 2-fold and 37 mature miRNAs down-regulated more than 2-fold in Toxoplasma gondii infected HFFs, and the validate result of 4 miRNAs level by quantitative realtime RT-PCR and Northern blot was consistent with the microarray data. Conclusion: Toxoplasma gondii infection modulates host miRNAs expression, and differential expressed miRNAs may play an important role on the interaction of the host- Toxoplasma gondii.
