乳香提取物对大鼠雪旺细胞增殖和相关蛋白表达的影响

摘　　要：通过研究乳香提取物对周围神经损伤的修复作用,为中药治疗创伤提供理论依据。将大鼠雪旺细胞株进行传代培养,通过MTT法筛选出乳香提取物干预大鼠雪旺细胞生长的安全浓度。在安全浓度范围内,将雪旺细胞随机分为对照组和药物组,用不同浓度的乳香提取物干预细胞生长,培养后第12、24、48、72和96小时,观察其对细胞增殖的影响;用流式细胞术检测细胞周期及DNA含量百分率;用ELISA试剂盒检测雪旺细胞分泌脑源性神经营养因子(BDNF)、神经细胞黏附分子(NCAM)及增殖细胞核抗原(PC-NA)表达量的变化。结果表明:乳香提取物干预大鼠雪旺细胞生长的安全浓度范围为62.5～8 000mg/L;当乳香提取物质量浓度为125、250和500mg/L时,与对照组相比增殖作用差异显著;MTT比色法及流式细胞仪检查结果证实125mg/L的乳香提取物促进雪旺细胞增殖的作用极为显著;乳香提取物在适当范围内可促进雪旺细胞增殖,不同质量浓度的促增殖能力不同,以125mg/L的效果最佳,并可使体外培养雪旺细胞内BDNF、NCAM、PCNA表达上调。Through studying the influence of frankincense extract on repairing peripheral nerve injury, thus offering the theoretical foundation for wound healing of herbs,rat Schwann cells（SCs） were subcultured as a experimental model, and it was cultured with different concentrations of the extract. In the range of safe concentration, SCs were randomly divided into control group and drug group,and different concen- tration of the extract was added to intervene cell growth. The cell proliferation was observed at hour 12, 24,48,72 and 96 post-cultivation. Flow cytometry method was applied to test cell cycle and the percent content of DNA. ELISA kit was used to detect the content of brain derived neurotrophic factor（BDNF）, neural cell adhesive molecule（NCAM） and proliferative cell nucleus antigen（PCNA）. Results showed that the safe concentration of the extract was from 62.5 mg/L to 8 000 mg/L. Compared with the control group, cell proliferation at the concentration of 125,250 and 500 mg/L increased significantly（P^0.05）. The re- sults of MTT colorimetric test and flow cytometry method showed that the extract could significantly pro- moted cell proliferation at 125 mg/L（P%0.01）. It suggested that the extract at certain concentration was effective on inducing the proliferation of SCs,and the best concentration was 125 mg/L. At the same time, it could up-regulate the expression of BDNF,NCAM and PCNA of SCs cultured in vitro.
