摘  要:采用二步降温法对匙吻鲟精子进行了超低温冷冻保存试验。分析比较了7种不同配比浓度稀释液、3种冻前低温平衡时间(0、30、50min)和3种冷冻平衡时间(5、10、20min)对匙吻鲟精子超低温冷冻保存效果的影响。试验结果表明,采用稀释液配方3(氯化钾0.5mmol/L,三羟甲基氨基甲烷20mmol/L,葡萄糖80mmol/L,pH 8.24)与精液按体积2∶1的比例稀释混合,加入8%甲醇,在2~4℃条件下平衡0~30min,分装1.5mL离心管,至液氮面2~3cm处平衡20min后投入液氮保存2h,38℃水浴解冻,用井水激活,获得最好解冻后活力为(35.95±4.77)%。The impacts of seven concentrations of diluent, three pre-freezing balance time(0 min, 30 min and 50 rain) and three frozen equilibrium time(5 min, 10 min and 20 min) on sperm cryopreservation were studied in sperm of paddlefish(Polyodon spathula) by two-step cooling program. The results showed that the best sperm cryopreservation with sperm motility of (35. 95±4. 77)M was obtained by the following procedure:the dilution NO. 3( KCI 0. 5 mmol/L, Tris-HC1 20 mmol/L, D-Glucose 80 mmol/L, pH 8. 24) and the semen were mixed at a ratio of 2 - 1 in volume and added 8% methanol, with the equilibration time within 30 min at 2-4 ℃. Then the dilution NO. 3 and the semen mixture were put into centrifugal tubes with capacity of 1.5 mL and soaked in the liquid nitrogen and kept for 2 hours after equilibration of 20 min at the position of 2-3 cm up the liquid nitrogen surface. The cryopreserved sperm was thawed in water of 38 ℃ and activated with well water.
