摘 要:【目的】探讨益气健脾中药党参对胃肠黏膜损伤修复的药效机理及物质基础。【方法】在水浸束缚应激性溃疡大鼠模型上观察党参水提液对胃肠黏膜损伤修复的作用;在小肠上皮细胞(IEC-6)上观察党参糖提取物对细胞迁移和多胺含量的影响。采用划痕法复制细胞迁移模型,柱前衍生—高效液相色谱法检测多胺含量。【结果】党参水提液能减轻应激性溃疡大鼠胃黏膜损伤,减轻病理损害,提高胃黏膜多胺(精脒)含量。党参糖提取物能促进IEC-6细胞迁移,逆转由二氟甲基鸟氨酸(DFMO)或4-氨基吡啶(4-AP)所致的细胞迁移抑制,并提高细胞迁移过程中细胞内多胺(精脒)含量;同时,可逆转由DFMO所致的多胺(精脒)含量降低。【结论】党参的胃肠黏膜损伤修复作用机制可能与其促进上皮细胞迁移,作用于多胺调控信号通路有关。Objective To investigate the pharmacodynamic mechanism of Radix Codonopsis,an herb with the actions of replenishing qi and strengthening spleen,for repairing gastrointestinal mucosal injury.Methods The stress ulcer rat model was established by water-immersion restrain for evaluating the repairing effect of Radix Codonopsis water extract on gastrointestinal mucosal injury.The effect of Radix Codonopsis polysaccharide on cell migration and polyamine(spermidine) content was observed in the intestinal epithelial cells(IEC-6).Cell migration was tested on IEC-6 migration model induced by scratch method,and T polyamines(spermidine) content was detected by precolumn derivatization-high performance liquid chromatography(HPLC).Results Radix Codonopsis water extract could relieve gastric mucosal injury in rats with stress ulcer,reduce pathological damage,and increase gastric mucosal polyamine(spermidine)content.Radix Codonopsis polysaccharide promoted IEC-6 cell migration,reversed the inhibitory effects of difluoromethylornithine(DFMO)or 4-aminopyridine(4-AP)on cell migration,increased intracellular polyamines(spermidine)content during cell migration,and reversed the decrease of polyamine content caused by DFMO.Conclusion Radix Codonopsis can repair thegastrointestinal mucosal injury,which might be related with the promotion of cell migration through polyamine-dependent signaling pathway.
