摘  要:【目的】探讨大黄素抑制小鼠结肠癌发展与影响调节性T细胞(Tregs)迁徙的相关性。【方法】选用BALB/c小鼠随机分为阴性对照组、模型组和大黄素组(剂量为100 mg.kg-1.d-1),后2组采用右前肢腋窝皮下接种CT26细胞制备结肠癌模型;采用流式细胞仪检测Tregs在结肠癌小鼠外周血、引流淋巴结和肿瘤局部的相对比例,以及趋化因子受体CCR4在Tregs的表达量差异,并研究其与肿瘤大小的相关性。【结果】大黄素组的肿瘤质量显著低于模型组(P<0.05),抑瘤率达74%;大黄素组Tregs在肿瘤局部的比例及其CCR4+Tregs的比例显著低于荷瘤模型组(均P<0.01)。【结论】大黄素抑制小鼠结肠癌的发展与其影响Tregs的迁徙相关,其机制与大黄素降低了CCR4在Tregs上的表达有关。Objective To investigate the correlation of the inhibitory effect of emodin on regulatory T cells(Tregs) migration with the development of mice colon cancer.Methods BALB/c mice were randomized into negative control group,tumor-bearing model group and emodin(100 mg·kg^-1·d^-1)group.Mice except for the negative control group were given subcutaneous inoculation of CT26 cells into the right axillary fossa of the mice to induce colon cancer.Flow cytometry was used to detect the relative proportion of Tregs in peripheral blood,draining lymph nodes and tumor mass of mice,and was used to measure the expression of chemokine receptor CCR4 on Tregs.The correlation of the relative proportion of Tregs and CCR4 expression level with the tumor mass was analyzed.Results The tumor mass of emodin group was significantly lighter than that of the tumor-bearing model group(P〈0.05),and the tumor-inhibition rate was up to 74%.The percentage of Tregs in the tumor mass and that of Tregs with positive CCR4 expression in the emodin group were significantly less than those in tumor-bearing model group(P〈0.01).Conclusion The results suggest that the mechanism of emodin in inhibiting the development of mice colon cancer should be related to the reduction of Tregs migration by down-regulating CCR4 expression on Tregs.
