摘  要:【目的】从感染了青枯病的广藿香植株中分离鉴定青枯菌,并进一步探讨青枯菌的寄主专化性。【方法】采用组织浸泡划线法分离青枯菌;选用青枯菌的特异性引物,对分离菌株进行PCR鉴定;以广藿香、番茄与花生为材料,通过离体叶片接种法和苗期接种法,进行寄主专化性研究。【结果】根据分离菌株在2,3,5-氯化三苯基四氮唑(TTC)培养基上的菌落特征、致病性回接实验和PCR鉴定结果表明,从感染了青枯病的广藿香植株中分离得到的菌株HX6、HX13、HX15与HX17为青枯菌Ralstonia solanacearum。对3种寄主的致病性结果表明,菌株HX15及HX17对广藿香的致病性明显高于番茄和花生;菌株HX6对3种寄主均有较强的致病性。番茄青枯菌GIM1.70对广藿香仅有弱致病性。【结论】从感染了青枯病的广藿香中分离鉴定了4个青枯菌菌株,菌株HX15及HX17的寄主专化性较强,菌株HX6的寄主专化性不明显。Objective To isolate and identify Ralstonia solanacearum from Pogostemon cablin(Blanco)Benth.infected with bacterial wilt,and to further explore their host specificity.Methods Ralstonia solanacearum strains were isolated by using streak plate method and were identified by polymerase chain reaction(PCR)with specific primers of Ralstonia solanacearum.With the plants of Pogostemon cablin,tomatoes and peanuts as the hosts,the host specificity of Ralstonia solanacearum strains was investigated by detached leaves inoculation method and seedlings inoculation method.Results Based on the results of colony characteristics on tetrazolium chloride(TTC) medium,and on the results of pathogenicity inoculation test and PCR identification,strains HX6,HX13,HX15 and HX17 isolated from Pogostemon cablin plants could be confirmed as Ralstonia solanacearum.Host specificity tests showed that strains HX15 and HX17 were more virulent on Pogostemon cablin than on tomatoes and peanuts,and strain HX6 had high pathogenicity to all of the three kinds of hosts.Strain GIM1.70 had weak pathogenicity to Pogostemon cablin.Conclusion Four Ralstonia solanacearum strains have been isolated and identified from Pogostemon cablin infected with bacterial wilt,strains HX15 and HX17 have high host specificity,while strain HX6 has low host specificity.
