摘 要:目的 观察中国汉族人群血管紧张素原(AGT)基因5个单核苷酸多态性(SNP)与2型糖尿病(T2DM)的相关性.方法 应用荧光标记单碱基延伸及寡核苷酸微阵列芯片杂交技术检测AGT基因rs3889728,rs2478523,rs1926722,rs7539020和rs2493132的基因型.结果 T2DM组rs2478523的CT基因型频率(0.382 vs 0.505)显著低于对照组(Z=5.664,P=0.017),而TT型频率(0.435 vs 0.307)显著高于对照组(Z=6.389,P=0.012).余4个SNP的基因型和5个SNP的等位基因频率两组间差异无统计学意义(Z=0.001~3.041,P值均〉0.05).根据连锁不平衡水平,由rs3889728,rs1926722和rs7539020构建的单倍型频率在两组间差异无统计学意义(P值均〉0.05).结论 AGT基因的rs2478523多态性可能是中国汉族人群T2DM的遗传易感因素.Objective To investigate the association between five single nucleotide polymorphisms (SNP) in angiotensinoge (AGT) gene with type 2 diabetes mellitus(T2DM) in Chinese Han nationality. Methods rs3889728,rs2478523,rs1926722, rs7539020 and rs2493132 of AGT gene were analyzed with allele-specific fluorogenic oligonucleotide probes in an assay combining extension and hybridization. Results In rs2478523, the frequency of CT genotype (0. 382 vs 0. 505, Z= 5. 664, P = 0. 017) was lower and TT genotype (0. 435 vs 0. 307, Z= 6. 389, P= 0. 012) was higher in T2DM group than control group. The frequencies of the other 4 SNPs' genotypes and all 5 SNPs' alleles were not significantly different between the two groups (Z=O. 001~3. 041,all P^0.05). According the level of linkage disequilibrium,the frequencies of the haplotypes constructed with rs3889728, rs1926722 and rs7539020 were not significantly different between the two groups (all P〉 0. 05). Conclusion Polymorphism of rs2478523 in AGT gene may be a genetic susceptibility factor for T2DM in Chinese Han nationality.
