摘　　要：目的 建立肠道病毒71型分离培养的Hep-2细胞法,探索最佳培养条件.方法 采用Hep-2细胞分别接种肠道病毒71型核酸检测阳性的咽拭子标本30份,观察标本在不同培养条件下的细胞病变.结果 分离肠道病毒71型22株,分离率为73.33%,肠道病毒71型接种细胞在35℃培养7天后出现拉网、圆缩、脱落等现象.降低培养温度（35℃）可提高病毒分离率.结论 初步建立了肠道病毒71型分离培养的Hep-2细胞法.Objective To develop a method of isolation and culture of enterovirus 71 （EV71） in Hep-2 ceils and optimize the culture condition. Methods 30 specimens enterovirus 71 positive detected by fluorescent quantitative real-time PCR were inoculated in Hep-2 ceils under different culture conditions to observe cytopathic effect （CPE）. Results A total of 22 entero- virus 71 strains were obtained from 30 specimens,and the positive rate was 73.33~. Cytopathic effect of viral infection in Hep-2 cells was observed after incubated at 35＊（2 for 7 days. Lower temperature （35℃） could increase the rate of virus isolation. Conclusion The method of culture of nterovirus 71 in Hep-2 ceils was developed.
