摘  要:目的探讨α-硫辛酸(α-LA)对葡萄糖/葡萄糖氧化酶(DG/GO)所致大鼠心肌细胞损伤的作用。方法将体外培养的大鼠心肌细胞株H9c2分成对照组(A组)、α-LA组(B组)、DG/GO组(C组)和α-LA+DG/GO组(D组)。24h后,倒置显微镜观察细胞形态,MTT检测细胞生存率,Hoechst 33342染色检测细胞凋亡率。结果与A组相比,C组细胞形态上变小、变圆、皱缩,细胞存活率下降(P<0.01),细胞核固缩率增加(P<0.01);而D组上述指标较C组均有明显改善(P<0.01)。结论α-LA可显著减轻DG/GO引起的H9c2细胞损伤。Objective To investigate the effect of α-Lipoic acid(a-LA) on injury of myocardial cells induced by d-glucose/glucose oxidase(DG/CK)) in rats. Methods Rat myocardial cell line H9c2 was cultured in vitro and divided into four groups of A (normal control), B(treated with α-LA 300 μmol/L), C (treated with E)G 30 mmol/L and C4D 5 mU/ml) and D (pretreated with α-LA 300μmol/L followed by DG 30 mmol/L and GO 5 mU/ml). The cell morphology was observed under inverted microscope 24 h later. The rates of cell survival and apoptosis were detected by MTT and Hoechst 33342 staining, respectively. Results Compared with group A, H9c2 cells became small, round and corrugativus and the rate of cell survival was decreased (P〈0. 01), while rate of cell pycnosis was increased (P〈0. 01) in group C, which were all significantly improved in group D (P〈0. 01). Conclusion α-LA pretreatment can markedly relieve the injury of H9c2 cells induced by DG/GO.
