丹参酮ⅡA对大鼠环孢素性牙龈增生干预治疗的初步研究

摘　　要：目的:初步观察丹参酮ⅡA(TanshinoneⅡA,TSN)对大鼠环孢素A(cyclosporinA,CsA)性牙龈增生的干预治疗效果。方法:90只7周龄雄性Wistar大鼠随机等分为3组(对照、CsA、CsA+TSN),每组再随机等分为3个亚组(10d、20d、30d)。Nikon 80i显微镜下,利用成像分析系统摄片并测量各组大鼠颊侧牙龈上皮面积和结缔组织面积,两因素析因设计方差分析测量结果。结果:与对照组相比,CsA组颊侧牙龈上皮面积和结缔组织面积均显著增大(P<0.01)。与CsA组相比,CsA+TSN组颊侧牙龈上皮面积和结缔组织面积均显著减小(P<0.01)。与对照组相比,CsA+TSN组的10d和20d亚组颊侧结缔组织面积均减小(P<0.05,P<0.01)。CsA组大鼠颊侧附着龈近膜龈联合处牙龈增厚明显,胶原纤维束明显增粗,胶原束间隙明显减小;CsA+TSN组大鼠牙龈形态与对照组相似,胶原纤维比CsA组明显变细,胶原束间隙明显增大。结论:TSN能够缓解CsA所致的大鼠牙龈上皮和结缔组织部位特异性的增厚,对结缔组织增生的缓解作用可能与其减少胶原沉积有关。Objective： To observe the preliminary effect of tanshinone IIA on the cyclosporine A –induced gingival overgrowth of rats.Methods： Ninety male Wistar rats of 7 weeks old were randomly divided into three groups（control,CsA,CsA＋TSN）,each group was further divided into three groups（10 days,20 days,30 days）.Serial paraffin sections were cut in a bucco-lingual direction throughout the mandibular first molars.Under the Nikon 80i microscope,the area of buccal gingival epithelial and connective tissue were measured with the image analysis system and the measurements were analyzed by two-way analysis of variance of factorial design.Results： The area of buccal gingival epithelial and connective tissue of the CsA group rats were significantly greater than the control group ones＇（P0.01）.The area of buccal gingival epithelial and connective tissue of the CsA＋TSN group rats were decreased significantly than the CsA group ones＇（P0.01）.The area of buccal connective tissue of the CsA＋TSN 10 days and 20 days group rats were decreased significantly than the control group ones＇（P0.05,P0.01）.The gingiva of CsA group rats approach muco-gingival junction were overgrowth obviously,the collagen were obviously thickening and collagen beam gap decreased significantly;while the gingival appearance of CsA＋TSN group rats were similar with the control group ones＇,and comparing to CsA group,the collagen of CsA＋TSN group rats were obviously thinning and collagen beam gap increased significantly.Conclusion： TSN can alleviate cyclosporine A-induced gingival epithelium and connective tissue regional specific overgrowth of rats,the effect for connective tissue may be associated with the reducing of collagen.
