雌酮、雌二醇、表皮生长因子和成纤维细胞生长因子对子宫内膜细胞体外增殖的影响

摘　　要：目的探讨雌酮(E_1)、雌二醇(E_2)、表皮生长因子(EGF)和成纤维细胞生长因子(FGF)对人子宫内膜腺上皮细胞和间质细胞体外生长的调控作用。方法对16例人子宫内膜离体组织进行体外分离培养,细胞汇合后消化,一部分用盖片法培养,用特异性抗体鉴定细胞纯度;一部分做细胞增殖试验,在相同的生长期分别加入指定浓度的E_1、E_2、EGF、FGF、E_2+EGF、EGF+FGF、E_2+FGF培养24、48、72 h,用四甲基偶氮唑盐微量酶反应比色法(MTT法)、流式细胞仪测定E_1、E_2、EGF、FGF对子宫内膜腺上皮细胞及间质细胞的作用。结果 2.5×10^(-5)mmol/L E_2、1.613 nmol/L EGF及0.617 nmol/LFGF刺激细胞生长,EGF+FGF刺激细胞生长的作用最强;2.5×10^(-5)mol/L E_2、1.613 nmol/L EGF、0.617 nmol/L FGF不刺激细胞凋亡;MTT法、流式细胞仪法结果一致。结论人子宫内膜腺上皮细胞和间质细胞的生长受到E_2、EGF及FGF的调控,E_2、EGF和FGF具有促进细胞增殖作用,EGF、FGF联合应用具有最佳的协同效应。Objective： To study the effect of estrone（E1 ）, estradiol-17 beta（E2）, epidermal growth factor（EGF） and fibroblast growth factor（FGF）on the proliferation of human endometrial stromal and epithelial cells. Methods： Sixteen samples of normal human endometrium were collected. The confluent cells were digested. Part of the cells was cultured by cover slip method, and the special antibody was used to identify the purity of the cell,while the rest cells were cultured with different concentrations of E1 ,E2 ,EGF,FGF, E2＋EGF,EGF＋FGF or E2 ＋FGF for 24,48 or 72 hours. The proliferation and apoptosis of cells were detected by MTT method and flow cytometry. Results： The cell growth speed was increased by 2.5× 10^-5 mol/L E2,1. 613 nmol/L EGF or 0. 617 nmol/L FGF（P〈0.01）,with the highest growth in EGF＋FGF group, but the apoptotic rates were not significantly different from those of control（P〉0.05）. Conclusions： E2, EGF or FGF can regulate the proliferation of human endometrial stromal and epithelial cells. E2. EGF or FGF significantly promote the proliferation of human endometrial cells at adefinite concentration. EGF and FGF can synergistically stimulate the proliferation of human endometrial ceils.
