摘  要:目的探讨姜黄素对脊髓缺血损伤后脂质过氧化诱发细胞凋亡的影响。方法建立新西兰大白兔脊髓缺血模型,分为缺血对照组、姜黄素给药组及假手术组。采用神经功能评分评价脊髓缺血兔后肢运动功能。采用硫代巴比妥酸(TBA)比色法、黄嘌呤氧化酶法、硝酸还原酶法、RT-PCR和流式细胞仪等技术检测丙二醛(MDA)、超氧化物歧化酶(SOD)、一氧化氮(NO)、半胱氨酸天冬氨酸蛋白酶3(Caspase-3)的表达水平和细胞凋亡率。结果与缺血对照组比较,姜黄素可降低MDA含量、NO含量和Caspase-3表达量,提高SOD活性,降低细胞凋亡率,差别有统计学意义。神经功能评分结果显示姜黄素给药组评分显著高于缺血对照组。结论姜黄素对兔脊髓缺血损伤具有保护作用,此作用与抑制脂质过氧化、从而减少缺血细胞凋亡有关。Objective To observed the effects of curcumin on apoptosis induced by lipid peroxidation after spinal cord ischemia. Methods Spinal cord isehemia model of New Zealand white rabbits were established,and then divided into ischemic control group, cureumin dose group and the sham-operation group randomly. Apply neurological score to evaluate the spinal cord ischemic rabbit hindlimb function. Apply thiobarbituric acid, xanthine oxidase, nitrate reductase method, RT-PCR and flow cytometry to detect the concentration of malondialdehyde(MDA), superoxide dismutase(SOD), nitric oxide(NO), the expression levels of caspase 3 and apoptosis rate. Results Compared to ischemic control group, curcumin can reduce the concentration of MDA, NO and Caspase-3 expression levels, increase SOD activity, reduce the rate of apoptosis, the difference was statistically significant. The neurological score results show that curcumin treatment group scores were significantly higher than the ischemic control group. Conclusion This study showed that curcumin has a protective effect on spinal cord ischemie injury, which may be related to its inhibition of lipid peroxidation, thereby reducing the ischemic cell apoptosis.
