电针刺天井、清冷渊配合至阴点刺放血对Ⅰ型超敏反应模型大鼠IL-4、IFN-γ表达水平影响随机平行对照研究

摘　　要：[目的]观察电针刺天井、清冷渊配合至阴点刺放血对Ⅰ型超敏反应模型大鼠IL-4、IFN-γ表达水平影响。[方法]使用随机平行对照方法,将40只健康雌雄各半SD大鼠按随机数字表随机分为4组(10只/组),空白对照组、模型组、电针组、阳性药物组。空白对照组不予处理,模型组按10mL/Kg.d予生理盐水灌胃。第6d开始,阳性药物组予等体积顺尔宁悬浊液灌胃干预,电针组针刺天井、清冷渊配合至阴点刺放血,连续灌胃及电针刺14d。药物与电针干预14d后,用手术剪取下以激发注射部位为中心的1cm×1cm皮肤组织,用免疫组化法测定皮肤组织中IL-4、IFN-γ表达水平,通过录像回放统计大鼠搔抓次数。[结果]电针可减少大鼠搔抓次数(P<0.01),同时具有上调INF-γ表达水平从而抑制IL-4表达的作用(P<0.01),对比顺尔宁片在缓解瘙痒症状、恢复细胞因子分泌平衡方面有相似治疗作用(P>0.05)。[结论]电针能够有效干预造模大鼠Ⅰ型超敏反应,改善瘙痒症状,上调INF-γ表达水平从而抑制IL-4表达,与顺尔宁片比较在改善Ⅰ型超敏反应症状方面有相似作用;其机制可能为上调INF-γ表达水平从而抑制IL-4表达,恢复细胞因子分泌平衡,从而治疗Ⅰ型超敏反应性疾病。[Objective]Observe the influence of type Ⅰ allergic inflammation rats’IL-4、IFN-γ expression level treated by electric acupuncture tianjing、qinglengyuan and zhiyin prick bleeded.[Method]40 healthy male and female half and half SD rats were randomly divided into 4 groups according to random number table with random parallel control method,10 rats every group,there were blank control group,model group,the electric acupuncture group,positive drug group.Bland control group rats were dealt with nothing,model group rats were gavaged with saline on the basis of 10mL/Kg.d.From the sixth day on,the positive drug rats were gavaged with the same volume of singulair suspension,the electric acupuncture group rats were acupunctured tingjing、qinglengyuan and zhiyin prick bleeded,gavaged and acupunctured 14 days.After 14 days of medicine and electric acupuncture intervention,cut down the stimulated injection site centered 1cm×1cm skin tissue,detected IL-4、IFN-γ expression level in the skin tissue by immunohistochemical method,counted the scratching frequency of the rats through the video playback.[Result]electric acupuncture can reduce the scratching frequency of rats（P0.01）,raise the expression level of INF-γand inhibit the expression level of IL-4（P0.01）,compared with singulair,they have similar effect on relieving itching symptoms and restoring cytokines secretion balance（P0.05）.[Conclusion]electric acupuncture can intervene type Ⅰallergic inflammation model rats effectively,it can relieve itching symptom,raise the expression level of INF-γand inhibit the expression level of IL-4,compared with singulair,they have similar effect on relieving the symptom of type I allergic inflammation;the mechanism may be through raising the expression level of INF-γand inhibiting the expression level of IL-4,restoring cytokines secretion balance,so as to treat type Ⅰ allergic inflammation disease.
