摘 要:目的:观察丹紫康膝冲剂对大鼠膝骨关节炎软骨端粒酶的影响,并探讨其防治膝骨性关节炎的作用机制。方法:健康Wistar大鼠80只,雌雄各半,随机分为丹紫康膝组、西药组、模型组、空白对照组,每组20只。对丹紫康膝组、西药组、模型组大鼠运用改良Hulth造模法进行大鼠膝关节骨性关节炎造模,于造模6周后对各组大鼠进行给药治疗。丹紫康膝组灌胃丹紫康膝冲剂,西药组灌胃硫酸氨基葡萄糖,模型组、空白对照组灌以等容积蒸馏水,每天早晚各1次,连续4周。肉眼观察大鼠膝关节大体形态,用TRAP-ELISA法检测关节软骨端粒酶活性。结果:肉眼观察下见模型组膝关节软骨明显退变,局部溃疡,关节边缘增生,丹紫康膝组和西药组可见膝关节有新生软骨生成,空白对照组膝关节软骨光滑润泽,丹紫康膝组和西药组软骨端粒酶活性较空白对照组及模型组高(P<0.05),丹紫康膝组和西药组软骨端粒酶活性之间差异无统计学意义(P>0.05)。结论:丹紫康膝冲剂能提高大鼠膝关节端粒酶活性,从而延缓大鼠膝骨性关节炎的进一步发展。To observe the effect of Danzi Kangxi granules on the activity of telomerase in articular cartilage among rats with knee osteoarthritis and to investigate the mechanism by which Danzi Kangxi granules prevent and treat knee osteoarthritis.Methods:Eighty healthy Wistar rats(male-to-female ratio = 1:1) were randomly divided into Danzi Kangxi group(n = 20),Western medicine group(n =20),model group(n = 20),and blank control group(n = 20).A rat model of knee osteoarthritis was established in the Danzi Kangxi group,Western medicine group,and model group by improved Hulth method.At 6 weeks after the model was established,the Danzi Kangxi group received Danzi Kangxi granules by gavage,the Western medicine group received glucosamine sulfate by gavage,the model group and blank control group received equal volumes of distilled water.Treatment was provided once in the morning and once at night for 4 consecutive weeks.The gross morphology of knee joints was observed under the naked eye.The activity of telomerase in articular cartilage was measured by TRAP-ELISA.Results:Under the naked eye,the model group showed significant degeneration of knee articular cartilage,with local ulcer and joint edge hyperplasia;the Danzi Kangxi group and Western medicine group had new cartilage formation in knee joints;the blank control group had smooth knee articular cartilage.The Danzi Kangxi group and Western medicine group had significantly higher activity of telomerase in articular cartilage than the blank control group and model group(P 0.05).There was no significant difference in activity of telomerase in articular cartilage between the Danzi Kangxi group and Western medicine group(P 0.05).Conclusion:Danzi Kangxi granules can improve the activity of telomerase in knee joints among rats,thus delaying the progression of knee osteoarthritis.
