摘  要:目的观察抵当芪桂汤对STZ致糖尿病大鼠晶状体、晶状体上皮细胞以及HSP70水平的影响。方法随机抽取10只SD大鼠为空白对照组,其余大鼠一次性腹腔注射链脲佐菌素(STZ)60 mg/kg,复制糖尿病大鼠模型。选取成模动物55只随机分为模型组、达美康组、抵当芪桂汤组、抵当芪桂汤大剂量组、抵当芪桂汤加达美康组,分别给予相应干预。于实验2、4、6、8 W在裂隙灯下检查晶体混浊情况;8 W后显微镜下察晶状体上皮细胞形态变化,并检测晶状体上皮细胞(LEC)中的HSP70表达情况。结果空白对照组大鼠晶状体始终保持透明完整,模型组大鼠晶状体逐渐以致完全混浊,其余治疗组大鼠晶状体混浊程度由重到轻依次为抵当芪桂汤大剂量组、抵当芪桂汤组、达美康组、抵当芪桂汤加达美康组;HE染色晶状体上皮细胞形态亦发生不同改变;以空白对照组大鼠作为标准,模型组晶状体LEC中HSP70表达逐渐增多,治疗组抵当芪桂汤大剂量组HSP70的阳性表达率高于达美康组、抵当芪桂汤组、抵当芪桂汤加达美康组HSP70的阳性表达率,差异具有显著性(P<0.05)。结论抵当芪桂汤通过降低血糖、减少糖代谢产物在晶状体内聚集、降低晶状体内渗透压从而有效保护实验性糖尿病大鼠晶状体损伤,抑制糖尿病大鼠白内障的发生和发展,对早期糖尿病大鼠白内障具有一定的防治作用。Objective To observe the effect of Didang Qigui decoction induced diabetic rats lens, lens epithelial cells and the level of HSP70 effects on STZ. Methods a total of 10 SD rats into normal control group, the other rats by intraper- itoneal injection of streptozotocin (STZ) 60 mg/kg, the model of diabetic rats. Selected into the model animal and 55 rats were randomly divided into model group, gliclazide group, Didang Qigui decoction group, Didang Qigui decoction high dose group, Didang Qigui decoction and Diamicron group, to give the corresponding intervention respectively. In Experi- ment 2, 4, 6, 8 W under slit lamp examination the opacity of the lens epithelial cells; morphology changes of lens to ob- serve 8W under the microscope, and the detection of lens epithelial cell (LEG) expression in HSP'/0. Results the blank control group rat lens remain transparent integrity, lens model group rats gradually so completely cloudy, the rest of the rats in the treatment group lens opacity degree from heavy to light are Didang Qigui decoction high dose group, Didang Qigui decoction group, giiclazide group, Didang Qigui decoction and Diamicron group; HE staining morphology of lens epithelial cells also have different change; in the blank control group rats were used as standards, gradually increased theexpression of HSP70 in LEC model group lens, the treatment group of Didang Qigui decoction large doses of the positive expression rate of HSP70 was high on the Hong group, arrived when the positive expression of Didang Qigui decoction group, Didang Qigui decoction and Diamicron group HSP'/0 the rate, the difference was significant ( P 〈 0. 05 ). Conclusion Didang Qigui decoction by lowering blood sugar, reducing sugar, reduce the accumulation of metabolites crystalline and osmotic pressure in vivo so as to effectively protect the lens damage in experimental diabetic rats in the crystalline body, and inhibit the development of cataract in diabetic rats, has a preventive effect on early diabetic rat cata- rac
