摘 要:目的:研究α-甘草次酸衍生物(GB)对大鼠外周血淋巴细胞体外活化的影响,探讨其是否对淋巴细胞活性存在抑制作用。方法:无菌分离大鼠外周血淋巴细胞并制成悬液,分别与不同浓度的GB预先孵育2 h,加入促分裂原伴刀豆蛋白A(ConA),继续孵育24 h后,ELISA法检测上清液中IL-4、IFN-γ含量。结果:2 mg·L^(-1)和1 mg·L^(-1)GB能显著降低ConA刺激下大鼠淋巴细胞产生的IL-4水平,而0.5 mg·L^(-1)GB则提高淋巴细胞产生的IL-4水平;2 mg·L^(-1)GB显著降低ConA刺激下大鼠淋巴细胞产生的IFN-γ水平,而1 mg·L^(-1)和0.5 mg·L^(-1)GB则提高淋巴细胞产生的IFN-γ水平。结论:给药剂量不同,GB对活化的大鼠外周血淋巴细胞作用不同,值得深入研究。To investigate the effects of a- glycyrrhetinic acid derivative(GB) on in vitro activation of peripheral blood lymphocytes in rats,and on inhibition of the activity of lymphocytes.Method:Lymphocytes suspension was isolated and prepared sterilely from peripheral blood in rats,and was incubated with different concentrations of GB for 2h,then polyclonal activator ConA was added to activate the lymphocytes.After 24h,IL-4 and IFN-γproduction of lymphocytes were determined by enzyme linked immunosorbent assay (ELISA).Result:It was found that GB(2,1 mg·L^(-1) )could significantly down-regulate IL-4 productions of lymphocytes in rats by ConA stimulation and GB(0.5mg·L^(-1) ) could up-regulate IL-4 production.Meanwhile GB(2 mg·L^(-1)) could significantly down-regulate IFN-γproductions of lymphocytes in rats by ConA activation and GB(1,0.5 mg·L^(-1) ) could up-regulate IFN-γproductions.Conclusion: With different dose,GB can exert different action on the peripheral blood lymphocytes in rats,which is worthy of further study.
