摘　　要：目的:建立测定人血浆中去甲万古霉素浓度的LC-MS/MS分析方法,用于临床去甲万古霉素血药浓度监测。方法:色谱柱为Shimadzu(VP-ODS 2.0 mm×150 mm,5μm),流动相为0.1%甲酸水溶液和乙腈,采用梯度洗脱,流速0.3 ml·min^(-1)。以罗哌卡因为内标,50μl血浆加入加30μl的25%(w/v)三氯乙酸沉淀蛋白并经离心处理后,上清5μl进样分析。结果:线性范围为0.5～100μg·ml^(-1),最低定量限为0.5μg·ml^(-1);日内、日间相对标准差<15%,相对误差为-4.1%～3.0%;提取回收率为79.8%～95.1%。结论:本方法样品简单、准确、灵敏度高,适用于去甲万古霉素临床常规血药浓度的监测。To establish a method for the determination of norvancomycin in human plasma by LC-MS/MS.Method： The separation was achieved on a Shimadzu column（VP-ODS 2.0 mm×150 mm,5μm）,the mobile phase consisted of 0.1% formic acid water solution and acetonitrile with gradient elution at a flow rate of 0.3 ml·min.Ropivacaine was used as the internal standard. After 50 pi plasma was precipitated with 30μl 25%trichloroacetic acid（w/v）,5μl supernatant was injected into the column for the analysis.Result：Linear calibration curves were obtained within the concentration range of 0.5-100μg·ml^（-1） with the lower limit of quantification of 0.5μg·ml^（-1）.The intra- and inter-day precision（RSD） values were below 15%and the relative error was from - 4.1%to - 3.0%at all QC levels.The extraction recovery was from 79.8%to 95.1%.Conclusion：The determination method for norvancomycin is convenient,accurate and sensitive,and can be applied in the analysis of norvancomycin in human plasma.
