摘 要:目的:建立测定人血浆中丙泊酚浓度的高效液相色谱荧光检测法。方法:血浆样品经甲醇沉淀蛋白后,取上清液进样分析,色谱条件为采用SymmetryC_(18)柱(3.9 mm×150 mm,5μm),甲醇-水(70:30)为流动相,流速1.0 ml·min^(-1),荧光检测激发波长276 nm、发射波长310 nm,进样量为20μl。结果:丙泊酚血药浓度在0.010~10.00μg·ml^(-1)内,线性关系良好(r=0.999 9),最低定量浓度为0.010μg·ml^(-1),提取回收率为93.41%~96.94%,方法回收率为98.65%~102.17%,日内、日间RSD均小于10%。结论:本方法快速、准确、灵敏、专一性好,适用于临床丙泊酚血药浓度检测和药动学研究。To establish a sensitive method for the determination of propofol in human plasma by HPLC with fluorescence detection.Method:The plasma samples were pretreated with methanol as the protein precipitation agent and separated on a Symmetry C_(18) column(3.9 mm×150 mm,5μm) with the mobile phase of methanol-water(70:30) at a flow rate of 1.0 ml·min^(-1),and the fluorescence detector was set at 276/310 nm.Result:A good linearity was obtained over the range of 0.010-10.00μg·ml^(-1)(r = 0.999 9).The extraction recovery was 93.41%-96.94%.Accuracy(the method recoveiy ) was 98.65%-102.17%.The inter- and intra-day RSD were both less than 10%.Conclusion:The method is rapid,sensitive and specific for the determination of propofol in human plasma in the clinical pharmacokinetic and drug monitoring studies.
