摘  要:目的:本研究旨在探讨培哚普利对慢性心力衰竭(CHF)患者外周血中CD4+CD25+Foxp3+调节性T细胞(Treg)表达失衡的调节作用。方法:选择CHF患者40例,在常规治疗基础上加用培哚普利4mg/d,治疗12周。在治疗前和治疗12周后采外周血标本,流式细胞学检测Treg占CD4+T细胞的比例,实时定量PCR检测外周血单个核细胞Foxp3mRNA的水平,体外增殖抑制试验检测Treg的抑制功能。结果:经过12周加用培哚普利的治疗后,患者Treg比例增加,Foxp3表达增高,并且Treg的抑制功能增强。结论:培哚普利通过上调Treg的表达,并恢复其抑制功能,调节CHF患者的免疫失衡而发挥其治疗作用。Objective:To investigate the modulation of Perindopril on imbalance of CD4+ CD25 + Foxp3 ~ Regu- latory T cell (Treg) in patients with chronic heart failure (CHF). Method:This study included 40 CHF patients and Perindopril was added to conventional therapy after they were admitted to our hospital. Blood samples were obtained before and 12 weeks after therapy. The proportion of Treg in CD4+ T cells was determined by flow cytometry, Foxp3 mRNA of Peripheral blood mononuclear cells was detected by Real-time PCR, and suppressive function of Treg was evaluated by in-vitro suppression assay. Result: After 12 weeks therapy of Perindopril, the proportion of Treg, expression of Foxp3 and suppressive function of Treg were all significantly increased compared with before. Conclusion: Perindopril modulates immune imbalance in patients with CHF through upregulation of Treg and recovery of its suppressive function.
