摘  要:目的:分离纯化发酵液中的α-葡萄糖苷酶抑制剂,并对其结构进行初步鉴定。方法:通过预处理发酵液、活性炭脱色、中空纤维超滤、Sephadex G25葡聚糖凝胶层析以及Superdex Peptide 10/300GL预装柱层析等方法对发酵液中的活性抑制剂进行了分离纯化,通过质谱鉴定确定其分子量,结合紫外-可见光光谱扫描、傅里叶红外光谱扫描以及硅胶薄层层析对该物质的结构进行初步分析。结果:经分离纯化,该物质的高效液相色谱图呈单一对称峰,通过质谱确定其分子量为1 228.8,经初步鉴定该抑制剂应属于寡糖类物质。结论:提取了发酵液中的α-葡萄糖苷酶抑制剂,其分子量为1 228.8,目前未见有文献报道,为糖尿病新药研发奠定了基础。Objective:To isolate and determine of alpha -glucosidase inhibitor from fermentation liquid. Method:Separation and purifica- tion of alpha - glucosidase inhibitor from fermentation liquid through pretreatment, decolorization with active carbon, hollow fiber ultrafihra- fion ,Sephadex G25 chromatography and Superdex Peptide 10/300GL chromatography. Molecular was identified by mass spectrometry anal- ysis while structure was identified by UV -visible spectral scanning, infrared spectroscopy and silica gel thin layer chromatography. Result: The alpha - glucocidase inhibitor thus obtained homogeneous as shown by HPLC, then the result of purification of alpha - glucocidase inhibitor was effective. A molecular weight of 1 228. 8 was determined by mass spectrometric analysis and the structure showed like a kind of oligosaccharide after identification. Conclusion: Alpha - glucocidase inhibitor with a molecular weight of 1 228. 8 was purified from fermentation liquid,which had been not reported by document. The study could lay a foundation for new drugs of diabetes.
