摘 要:目的:探讨红景天(salidroside)对晚期氧化蛋白产物(AOPPs)诱导小鼠足细胞转分化的影响。方法:以小鼠永生性足细胞系为研究对象。给予200μg/ml的AOPP刺激小鼠足细胞24h,同时加入红景天(浓度分别为0.1μmol、1μmol、10μmol、100μmol、200μmol)进行干预,采用Westernblot检测内质网应激标志性蛋白Grp78、CHOP以及平滑肌激动蛋白(α-SMA)、nephrin、podocin的表达水平。结果:200μg/mlAOPP条件下小鼠足细胞表达高水平的Grp78、CHOP及α-SMA,表达低水平的nephrin和podocin;予不同浓度红景天作用后,足细胞中Grp78、CHOP及α-SMA的表达水平降低,neph-rin和podocin的表达水平升高。结果表明红景天可部分逆转AOPPs的作用,且存在一定的剂量依赖性。结论:红景天对AOPPs诱导的足细胞Grp78、CHOP过度表达有很好的抑制作用,这可能是它减轻肾脏纤维化的机制之一。Objective:To investigate the effect of Salidroside on Mouse Podocytes Epithelial-mesenchymal transition(EMT) induced by Advanced Oxidation Protein Products(AOPP).Methods:Using mice glomerular podocyte cell line as an in vitro system,podocytes were stimulated by AOPPs(200 μg/ml) and Salidroside(0.1 μmol,1 μmol,10 μmol,100 μmol,200 μmol)for 24 hours.Then the cells were collected and expression of alpha-smooth muscle actin(α-SMA),Grp78(glucose-regulated protein78),CHOP,nephrin and podocin were detected by Western blot.Results:Under AOPPs(200 μg/ml) condition,there were high expressions of Grp78,CHOP and α-SMA,and low expression of nephrin and podocin in mice podocyte cell.After the treatment with Salidroside(0.1 μmol,1 μmol,10 μmol,100 μmol,200 μmol),the expressions of Grp78,CHOP and α-SMA were decreased and the expression of nephrin and podocin were increased in mice podocyte cell.The results showed that Salidroside could partly reverse the effect of AOPPs in a dose-dependent manner.Conclusion:Salidroside can attenuate renal fibrosis,and the protective mechanism may be partly through by regulating the over expressions of genes Grp78 and CHOP.
