摘 要:目的:探讨中药复方糖肾宝(主要由黄芪、大火草、葛根、莪术、三七组成)对高糖培养的大鼠系膜细胞(MCs)增殖的影响及可能作用机制。方法:体外培养大鼠MCs,随机分为正常组、高糖对照组、糖肾宝低、中、高剂量组、厄贝沙坦组。除正常组外,其余各组分别予以30mmol/L葡萄糖+正常大鼠血清、糖肾宝低、中、高剂量血清、厄贝沙坦血清进行干预48h。MTT法检测细胞增殖,RT-PCR法检测单核细胞趋化蛋白-1(monocyte chemoattractant protein-1,MCP-1)、转化生长因子-β1(transforming growth factor-β1,TGF-β1)mRNA表达,ELISA检测MCP-1蛋白,western印迹检测TGF-β1蛋白表达。结果:高糖作用下的各组MCs增殖、MCP-1和TGF-β1表达均显著高于正常组(P<0.05,P<0.01);药物干预的各组MCs增殖、MCP-1和TGF-β1表达均显著低于高糖对照组(P<0.05,P<0.01);糖肾宝高剂量组MCs增殖、MCP-1和TGF-β1表达与厄贝沙坦组差异无统计学意义(P>0.05)。结论:中药复方糖肾宝能抑制高糖培养的MCs细胞增殖,其机制可能与抑制炎症反应有关。Objective:To investigate the effect of Tangshenbao Granule on the proliferation of rat glomerular mesangial cells(MCs) induced by high glucose and its mechanism.Methods:The rat MCs were randomly divided into normal group(normal rat serum),high glucose control group(30 mmol/L glucose and normal rat serum),Tangshenbao low(30 mmol/L glucose and serum containing low dose Tangshenbao),middle(30 mmol /L glucose and serum containing middle dose Tangshenbao) and high-dose(30 mmol /L glucose and serum containing high dose Tangshenbao) group,irbesartan group(30 mmol /L glucose and serum containing irbesartan).Cell proliferation was detected by MTT.TGF-β1 and MCP-1 mRNA were observed by real-time PCR.Expression of MCP-1 protein was examined by Enzyme-Linked Immunosorbnent Assay.Expression of TGF-β1 protein was examined by Western blot.Results:Compared with normal group,in all groups induced by high glucose the proliferation of MCs,MCP-1and TGF-β1 were significantly higher(P〈0.05,P〈0.01);Compared with high glucose control group,in all intervention groups the proliferation of MCs,MCP-1and TGF-β1 were significantly lower(P〈0.05,P〈0.01);Compared with irbesartan group,in Tangshenbao high-dose group the proliferation of MCs,MCP-1 and TGF-β1 expression was not significantly different(P〈0.05).Conclusion:Tangshenbao can inhibit the proliferation of MCs induced by glucose,and its mechanism may be related to inhibiting inflammatory reaction.
