青蒿酯对房颤大鼠心肌核转录因子-κB表达的干预作用

摘　　要：目的通过建立房颤大鼠模型,观察核转录因子-κB(nuclear factor kappa B,NF-κB)mRNA的表达水平以及青蒿提取物对其表达的影响。方法 40只大鼠按随机数字表法分为空白对照组、模型组、青蒿组、卡托普利组,每组10只。利用舌下静脉注射乙酯胆碱和氯化钙的方法建立房颤大鼠模型,使用青蒿提取物进行干预,以卡托普利作为对照药。实验前2周青蒿组以1.25g生药·kg-1灌胃,卡托普利组以3.25mg·kg-1灌胃,对照组及模型组均以等体积生理盐水灌胃。通过PCR以及Western-blot技术观察NF-κB mRNA表达以及NF-κB蛋白表达。结果与对照组相比,模型组大鼠心肌NF-κB mRNA的表达显著增加(P<0.05)。青蒿组能明显减少房颤大鼠心肌NF-κB mRNA的表达(P<0.05),卡托普利对NF-κB mRNA的表达无影响。NF-κB蛋白的表达与mRNA的表达呈一致规律。结论 NF-κB与房颤的发生关系密切。房颤大鼠NF-κB的表达明显增加。青蒿提取物能够有效地减少房颤大鼠心肌NF-κB的表达,这可能是青蒿抗心律失常的机制之一。Objective Through the establishment of a rat model of atrial fibrillation, observed the expression of NF-κB mRNA expression levels, and affect Artemisia annua extract has on it. NF-κB mRNA expression and protein expression were observed by PCR and Western-blot. Re- suits Compared with the control group, model group rat cardiac NF-κB mRNA expression was significant-ly increased （P 〈 0.05 ）. The Artemisia annua group could significantly reduce the atrial fibrillation rat myocardial NF-KB mRNA expression （P 〈 0. 05 ）. Captopril has no effect on the expression of NF-KB mR- NA. NF-KB protein expression and mRNA expression was consistent with the law. Conclusion NF-KB are closely related to the incidence of atrial fibrillation. Atrial fibrillation rat NF-KB expression was signifi- cantly increased. Artemisinic extract can effectively reduce fibrillation Myocardial of NF-KB expression, which may be one of artemisinic antiarrhythmic mechanism.
