肠道病毒71型感染对BALB/c乳鼠T淋巴细胞亚群的影响

摘  要:目的观察BALB/c乳鼠感染肠道病毒71型(EV71)后机体的免疫功能状态,为临床治疗提供依据。方法将30只1日龄BALB/c乳鼠随机分为对照组、实验组1组和实验2组,分别颅内注射20μL灭活EV71200804株、EV71200804株和EV71200803株;感染后7 d后引颈处死小鼠,取胸腺和脾脏,分离淋巴细胞进行T细胞亚群检测。结果与对照组比较,实验1组胸腺淋巴细胞中CD3+CD4+、CD3+CD8+和CD3+细胞百分比均明显升高(P均<0.05),实验2组各T淋巴细胞百分比略有升高(P均﹥0.05);实验1组脾脏淋巴细胞中CD+3CD+4、CD+3CD+8和CD+3细胞百分比均显著降低(P均<0.01),实验2组CD3+CD4+细胞百分比明显降低(P<0.05)、CD3+CD8+和CD3+细胞百分比无显著变化(P均﹥0.05);实验1组和实验2组胸腺CD4+ T细胞/CD8+T细胞均略低(P均﹥0.05),实验1组脾脏CD4+ T细胞/CD8+ T细胞略高、实验2组略低(P均﹥0.05)。结论 EV71感染可引起BALB/c乳鼠T淋巴细胞亚群功能紊乱,且其程度与病毒株有关。Objective To observe the immune function of enterovirus 71 ( EV71 )-infected neonatal BALB/c mice, which will provide basis for the clinical treatment. Methods Thirty one-day-old BALB/c mice were divided into 3 groups : the experimental group 1, experimental group 2 and the control group. The mice in the control group and experimental group were infected with 200804 and 200803 EV71 strains isolates respectively. All mice were sacrificed at 7d after the in- fection to collect the spleen and thymus tissues, and then the T-lymphocyte subsets were detected. Results Compared with the control group, the percentages of CD3+ CD4 , CD3+ CDs and CD3+ of thymic lymphocytes in the experimental group 1 were significantly increased ( all P 〈 O. 05 ), the percentages of T-lymphocytes in experimental group 2 were increased slightly ( all P 〉 O. 05) ; the percentages of CD3+ CD4, CD3 CD8 and CD3+ of the spleen lymphocytes in the experimental group 1 were significantly decreased ( all P 〈 O. O1 ), the percentage of CD3+ CD4 in the experimental group 2 was decreased significantly ( P 〈 O. 05 ), and no significant changes in CD3 CDs and CD3 ( all P 〉 O. 05 ) ; thymus CD4+ T cells and / CDs T cells in the group 1 and 2 were slightly lower ( P 〉 O. 05 ), spleen CD4 T cells/CD8 T cells in the group 1 was slight- ly higher, that in group 2 was slightly lower ( all P 〉 O. 05). Conclusion EV71 infection can cause disorders of T lym- phocyte subsets of neonatal mice, and its severity is related to the strains of the virus.
