摘  要:目的观察子痫前期(PE)患者外周血中CD4+CD25+调节性T(T)细胞及IL-10、TGF-β1的表达,分析其在PE发病机制中的作用。方法 PE患者28例(PE组)及正常晚期妊娠者28例(对照组),均收集外周抗凝静脉血,采用流式细胞仪检测外周血CD4+CD25+T细胞表达,计算CD4+CD25+T细胞占CD4+T细胞的比例;采用ELISA法检测血清IL-10、TGF-β1含量。结果 PE组和对照组CD4+CD25+T细胞分别为1.80%±0.26%、3.65%±0.57%,CD4+CD25+T细胞/CD4+T细胞分别为0.12±0.03、0.16±0.02,PE组两指标均显著低于对照组(P均<0.05);PE组和对照组血清IL-10分别为(28.30±13.29)、(42.10±12.85)pg/mL,TGF-β1分别为(180.54±81.52)、(253.75±93.41)pg/mL,PE组两种细胞因子含量均显著低于对照组(P均<0.05);PE组CD4+CD25+T细胞/CD4+T细胞与血清IL-10、TGF-β1均呈显著正相关,r值分别为0.52、0.62(P均<0.05),对照组CD4+CD25+T细胞/CD4+T细胞与血清IL-10、TGF-β1均无明显相关性。结论 PE患者外周血中CD4+CD25+T细胞表达及IL-10、TGF-β1均显著下降,细胞免疫抑制功能减弱及母胎免疫耐受失衡可能在PE发病中具有一定作用。Objective To observe the expression of CD4+CD25+5 Treg cells (T cells), IL-10 and TGF-β1 in peripheral blood of pre-eclampsia (PE) patients, and to analyze its role in immunopathogenesis of PE. Methods The peripheral blood from 28 hospitalized patients confirmed of PE ( PE group) and 28 cases of people with normal late pregnancy ( control group) was collected. The expression of CD4+CD25+ T cells was detected by flow cytometry, the ratio of CD4+CD25+T cell / CD4 + T cell was calculated, and the serum levels of IL-10, TGF-131 were detected by ELISA. Results The expression levels of CD4+CD25+T cells in the PE group and control group were 1.80% ± 0.26% and 3.65% ± 0.57% , respectively; the ratios of CD4+CD25+ T celL/CD4+ T cell in the PE and control groups were 0.12 ± O. 03 and O. 16 ± 0.02, respectively ; the two parameters of PE group were significantly lower than those in the control group ( all P 〈 0.05 ). The serum level of IL-10(28.30 ± 13.29) pg/mL was also decreased in PE group compared with control group (42.1 ± 12.85)pg/mL; the serum level of TGF-β1 ( 180.54 ± 81.52) pg/mL was also decreased in PE group compared with control group ( 253.75± 93.41 ) pg/mL( all P 〈0.05 ). In the PE group, CD4+CD25+ T cell/CD4 + T cell in the peripheral blood was positively corre- lated with the serum level of IL-10 and TGF-β1 (r =0.52, r = 0.62 respectively; all P 〈 0.05 ), but there was no signifi- cant correlation in the control group. Conclusion The expression of CD4+CD25+T cells and the serum levels of IL-10, TGF-β1 in the peripheral blood of PE patients are significantly decreased, cellular immunological function is suppressed and the materual-fetal immune tolerance is out of balance, which may be involved in PE immunopathogenesis.
