摘 要:以长白3号、东农427和龙稻9号成熟胚为试验材料,采用离体培养法研究培养基种类、激素种类和浓度以及Cu2+对水稻成熟胚再生体系的影响,对寒地粳稻再生体系进行优化。结果表明,长白3号和龙稻9号宜采用NMB+2,4-D 3 mg·L-1+6-BA 1.5 mg·L-1作为诱导培养基,而东农427宜采用NMB+2,4-D 3 mg·L-1+6-BA 1 mg·L-1作为诱导培养基,愈伤组织诱导率由优化前的49.00%、42.50%和68.01%分别提高到87.50%、91.30%和81.20%;东农427和龙稻9号宜采用NMB+0.02 mmol·L-1Cu2+培养基进行继代,长白3号宜采用NMB+0.01 mmol·L-1Cu2+培养基进行继代,愈伤组织褐化率分别由优化前的51.85%、29.41%和43.24%降低到23.61%、14.28%和21.60%;长白3号和龙稻9号宜采用N6+3 mg·L-16-BA+1 mg·L-1NAA+0.02 mmol·L-1Cu2+作为分化培养基,东农427宜采用N6+3 mg·L-16-BA+1 mg·L-1NAA+0.01 mmol·L-1Cu2+的N6培养基进行分化培养,愈伤组织分化率分别由优化前的12.90%、12.50%和2.38%提高到80.76%、46.15%和31.25%。To optimize the regeneration system of japonica rice in cold area, the effect of media and hormone types, hormone concentration and copper ions on the regeneration system of three japonica rice mature embryos were examined by in vitro culture. The results showed that the optimal medium for Changbai3 and Longdao9 induction was NMB+3 mgo L1 2, 4-D+1.5 mg. L1 6-BA, while NMB+3 mg. L1 2, 4-D+ 1 mg. L· 6-BA was for Dongnong427, and the induction rates of three rice mature embryos were increased from 49.00%, 42.50% and 68.01% to 87.50%, 91.30% and 81.20%, respectively. With the optimal subculture medium of NMB+0.02 mmol· L^-1 Cu^2+, browning rate of Dongnong427 and Longdao9 was decreased from 51.85% and 29.41% to 23.61% and 14.28%. But NMB medium containing 0.01 mmol· L^-1 copper ions was the best subculture medium for Changbai3, and the browning rate was decreased from 43.24% to 21.60%. The best suitable differentiation medium for Changbai3 and Longdao9 was N6+3 mg.L1 6-BA+I mg.L^-1 NAA+0.02 mmol.L-1 Cu^2+, however, N6 medium including 3 mg.L^-1 6-BA, 1 mg· L^-1 NAA and 0.01 mmol^ L^-11 Cu^2+ was the best optimal differentiation medium. The differentiation rates were raised from 12.90%, 12.50% and 2.38% to 80.76%, 46.15% and 31.25%.
