摘 要:目的探讨促红细胞生成素(EPO)对体外循环(CPB)大鼠肾脏组织核因子-κB(NF-κB)p65表达的影响。方法将40只雄性SD大鼠随机均分为4组(n=10):假手术组、模型组、低剂量EPO组和高剂量EPO组。测定血清肌酐水平,检测肾组织中NF-κB p65蛋白、基因表达,并行HE染色观察组织形态改变。结果模型组血清肌酐水平明显高于假手术组和EPO干预组(P<0.05),高剂量组低于低剂量组(P<0.05);与模型组相比,EPO组肾组织中NF-κB p65蛋白及基因表达水平显著下降;组织病理学检查显示EPO组的肾小管上皮细胞肿胀、胞浆内空泡形成、间质出血等病理变化较模型组明显减轻。结论 EPO抑制NF-κB p65的激活,减轻CPB术后肾脏损伤,并呈剂量依赖性。Objective To explore the effect of erythropoietin(EPO) on the expression of nuclear factor-kappaB (NF-κB) P65 in the renal tissue of rats undergoing cardiopulmonary bypass(CPB). Methods 40 male Sprague-Dawley rats were randomly divided into four groups : Sham, Control, Low and High EPO. Blood samples were collected for assays of serum creatinine at 24 h after the operation. Meanwhile renal samples were obtained for the determination of NF-κB P65 protein and gene, and for the histo- logic examination. Results Compared with Control group, the level of serum creatinine were lower in EPO groups (P 〈 0.05 ). EPO had effective inhibitory effect on the expression of NF-κB P65 protein and gene in renal tissues 24 h post-CPB(P 〈0.05), Histopathologic finding of the CPB group confirmed that there was renal impairment by cast formation and tubular necrosis in the tubular epithelium. Furthermore,EPO attenuated these above changes dose dependently. Conclusion EPO was effective on preventing CPB-induced renal injury probably through its upregulation of NF-κB P65.
