摘 要:目的阐明长春市2009年手足口病(Hand Foot and Mouth Disease,HFMD)流行期间,肠道病毒71型(Enterovirus Type 71,EV71)分离株的VP1编码区基因特征。方法对从长春市2009年HFMD患者分离到的7株EV71分离株,采用逆转录-聚合酶链反应(Reverse Transcription-Polymerase Chain Reaction,RT-PCR)进行VP1编码区基因扩增,并对扩增产物进行序列测定,用生物信息学软件Bioedit和Mega 4.0进行同源性、亲缘关系分析。结果 7株EV71分离株的核苷酸和氨基酸序列同源性分别为96.5%~100%和98.6%~100%;与安徽省阜阳市2008年流行株的核苷酸和氨基酸序列同源性分别为97.1%~99.7%和98.6%~100%。7株分离株均属于C4a基因亚型,在基因亲缘关系树上处于两个相对独立的进化分支(较大进化分支Clade 1和较小进化分支Clade 2)。结论目前监测到的2009年致长春市HFMD流行的EV71为C4a基因亚型,并且至少有2个C4a基因亚型的传播链同时存在,其中Clade 1为优势传播链。Objective To clarify genetic characteristics of VP1 coding region of enterovirus type 71 ( EV71 ) during epidemic period of hand foot and mouth disease ( HFMD ) in Changchun prefecture in 2009.Methods EV 71 strains were isolated from the patients of HFMD during the epidemic in Changchun in 2009.The VP1 coding region of 7 EV71 strains was amplified by reverse transcription-polymerase chain reaction ( RT-PCR ) .The RT-PCR products were sequenced and then the sequences were analyzed for homology and phylogenetics by Bioedit and Mega4.0.Result The identity of nucleotide and amino acid of these 7 strains was 96.5 %-100 % and 98.6 %-100 % respectively;There are 97.1 %-99.7 % nucleotide identity and 98.6 %-100 % amino acid identity compared with those epidemic strains in Fuyang city of Anhui province in 2008.All 7 strains were C4a sub-genotype and they formed two relatively separated clusters ( one was major cluster and the other was smaller one ) .Conclusions All 7 EV71 strains causing HFMD epidemic were C4a sub-genotype.There are at least two transmission chains caused HFMD epidemic in Changchun in 2009 with clade1 being the dominant transmission chain.
