3种石斛兰以茎段为外植体的离体快繁技术

摘　　要：以肿节石斛(Dendrobium pendulum)、尖刀唇石斛(D.heterocarpum)和报春石斛(D.primulinum)的无菌茎段为外植体,通过正交试验和单因子试验,研究不定芽诱导、生根和炼苗移栽等技术,筛选出各培养阶段的适宜培养基配方和培养条件,建立3种石斛兰离体快繁体系。结果表明:适合肿节石斛和报春石斛不定芽诱导的培养基为1/2MS+6-BA 0.5 mg.L-1+NAA 0.1 mg.L-1+椰乳100 mL.L-1,诱导率分别为93.75%和90.00%;适合尖刀唇石斛不定芽诱导的培养基为1/2MS+6-BA 0.25 mg.L-1+NAA 0.1 mg.L-1+椰乳100 mL.L-1,诱导率达到95.00%。适合肿节石斛和尖刀唇石斛生根的培养基为MS+NAA 0.5 mg.L-1+香蕉泥100 g.L-1,适合报春石斛生根的培养基为MS+NAA 0.75 mg.L-1+土豆泥100 g.L-1,生根诱导率高达100%。An in vitro propagation system of Dendrobium pendulum, D. heterocarpum and D. primulinum was established with sterile stems as explants by single factor and orthogonal experimental design. Shoot multiplication and rooting were investi- gated to screen perfect mediums and culture conditions. The best shoot multiplication medium of D. pendulum and D. primulinum is 1/2MS medium supplemented with 0.5 mg · L^-1 6-BA, 0.1 mg · L^-1NAA and 100 ml · L^-1CW, and the inducing rates are 93.75% and 90.00%, respectively. The best shoot multiplication medium of D. heterocarpum is 1/2MS me- dium supplemented with 0.25 mg · L^-1 6-BA, 0. 1 mg · L^-1 NAA and 100ml · L^-1 CW, and the inducing rates were 95.00%. The best rooting medium of D. pendulum and D. heterocarpum is MS medium supplemented with 0.5 mg · L^-1 NAA and 100 g · L^-1 Banana Juice. The best rooting medium of D. primulinum is MS medium supplemented with 0.75 mg · L^-1 NAA and 100 g · L^-1 mashed potato with the rooting rate of 100%.
