摘 要:[目的]为了分离鉴定吊兰附生甲基营养菌。[方法]通过微宇宙方法对吊兰进行甲醇和甲醛喷施和熏蒸30 d,采用水洗、过滤和离心从吊兰叶际和根际采样;优化的甲基营养菌无机盐筛选培养基中添加1%(V/V)甲醇和0.1%甲醛(V/V)混合液为仅有碳源,并添加50 mg/L放线菌酮抑制真菌生长;结合16S rDNA技术,鉴定菌株类型。[结果]分离鉴定12株甲基营养菌,其中叶际分离甲基营养菌属菌株和根际分离分支杆菌属菌株分别显示出叶、根代表性甲基营养菌特征,可分别耐受0.1%(V/V)和0.3%(V/V)的甲醛。[结论]该研究建立了吊兰附生甲基营养菌的一种有效分离和鉴定方法。[Objective] The research aimed to identify the epiphytic formaldehyde-tolerant methylotrophic bacteria from Chlorphytum comosum.[Method] Chlorphytum comosum leaves and roots were first collected using water-washing from C.comosum,and then potential methylotrophs were screened by an optimized nutrient medium added with 1%(V/V) methanol and 0.1%(V/V)formaldehyde as the only carbon source using 50 mg/L cycloheximide for inhibiting the fungal growth,and those isolated bacteria strains were further identified by the 16S rDNA technique.[Result] Twelve strains of methylotrophic bacteria were characterized that they could tolerate 0.1%-0.3% formaldehyde,wherein half of them belonged phyllosphere-derived Methylobacterium and rhizosphere-derived Mycobacterium,and they could tolerate 0.1% and 0.3% formaldehyde,respectively.[Conclusion] The study had successfully establish a method for screening and identifying C.comosum phyllospheric and rhizospheric methylotrophic bacteria.
