荷胆胶囊大鼠长期毒性实验

摘　　要：目的观察荷胆胶囊对SD大鼠的长期毒性。方法 SD大鼠100只,随机分为空白对照组和荷胆胶囊高、中、低剂量组,每组25只。荷胆胶囊各剂量组分别按1 600,800,400 mg.kg-1.d-1灌胃给予荷胆胶囊内容物;空白对照组灌胃给予0.9%氯化钠溶液,10 mL.kg-1.d-1。均连续给药30 d,停药次日每组取大鼠16只处死并检测血常规、血液生化、脏器系数和脏器组织病理,剩余大鼠停药2周,次日处死并检测血液学指标和病理情况。结果与空白对照组比较,荷胆胶囊各剂量组大鼠摄食量、体质量增长无明显差异,血液学、生化指标和脏器系数无明显变化,病理检查均未见与药物相关的明显病变,停药后未见药物延迟毒性反应。结论荷胆胶囊大鼠长期毒性实验未见异常反应。Objective To observe toxic reaction of SD rats after 30 days of hedan capsule administration to study the toxicity and evaluate the safety of the drug. Methods One hundred SD rats were randomly divided into 4 groups. Three groups of rats were intragastrically administrated with contents of hedan capsule at 1 600,800 and 400 mg ·kg-1 · d-1 , respectively. The control group was administrated with 0.9% NaCl at 10 mL ·kg-1 · d-1. The length of administration period was 30 days. Sixteen rats were executed after the cessation of administration, and the rest were executed 24 h after the two weeks recovery period. Routine blood test and blood biochemical test were performed. The organ coefficients were checked. Results Food intake and weight of rats in drug groups had no significant difference from those of the control group. The physical appearance and behavior, organ coefficients, the results of routine blood test and blood biochemical test of rats were not significantly different between the drug groups and the control group. Pathological examination discovered no obvious changes related to acute or delayed toxicity reactions after drug administration.Conclusion There are no abnormal changes in chronic toxicity test of hedan capsule in rats.
