摘 要:利用胡桃楸土壤中莽草酸的高效液相色谱测定方法,考察同一样地不同时期胡桃楸根际土和非根际土的莽草酸的含量。样品用乙酸乙酯水浴提取,蒸干并溶解于甲醇中。流动相为甲醇和1%磷酸水溶液(15∶85),流速1 mL/min,色谱柱为Agilent ZORBAX SB-C18(4.6 mm×150 mm,5μm),检测波长213 nm,柱温25℃。莽草酸在3.2~44.8μg/mL范围内与峰面积呈良好的线性关系(r=0.999 8),平均加样回收率为97.85%,RSD为0.69%(n=6)。胡桃楸根际土莽草酸含量大于非根际土中的含量,并且随着时间的推移莽草酸的含量出现先增长、后降低的变化。This study investigated manchurian walnut(Juglans mandshurica Maxim) rhizosphere and bulk soils at the same plot in different periods to establish the determination method for shikimic acid in manchurian walnut soils by high performance liquid chromatography(HPLC).Samples were extracted with ethyl acetate by water-bath heating,then evaporated and dissolved in methanol,and finally carried out separation and detection by HPLC.The mobile phase was methanol-1% H3PO4(15: 85);the flowing velocity was lml/min;Column separation was Agilent ZORBAX SB-C18(4.6mm × 150mm,5μm).The test wavelength was 213nm and column temperature was 25℃.The results Showed that shikimic acid in 3.2-44.8μg/mL range showed good linear relationship with peak area(r = 0.9998).The average recovery rate of shikimic acid was 97.85% and RSD = 0.69%(n = 6).The content of shikimic acid in manchurian walnut rhizosphere soils was higher than bulk soils,and the content of shikimic acid increased at first,then decreased by the change of time.
