温补肾阳汤对哮喘小鼠EOS分化的干预作用

摘　　要：目的:探讨温补肾阳汤对哮喘小鼠骨髓悬液EOS计数、血清白细胞介素-5(IL-5)和肺组织干细胞因子(SCF)表达的影响。方法:健康雌性小鼠50只,随机分为5组:正常组、模型组、醋酸泼尼松组、温补肾阳汤组、温补肾阳汤+醋酸泼尼松组,每组各10只。建立小鼠哮喘模型,各组分别于实验第29天开始给予药物灌胃,每天1次,共14天。计数骨髓悬液EOS;光镜下观察肺组织病理学改变;ELISA检测血清IL-5表达;免疫组织化学方法检测肺组织SCF蛋白表达。结果:与模型组比较,3个治疗组小鼠骨髓悬液EOS计数明显减少(P<0.01),外周血IL-5及肺组织SCF表达明显降低(P<0.01);与温补肾阳汤组比较,温补肾阳汤+醋酸泼尼松组与醋酸泼尼松组小鼠的骨髓悬液EOS计数、外周血IL-5及肺组织SCF表达均有下降(P<0.05);温补肾阳汤+醋酸泼尼松组与醋酸泼尼松组比较,差异无统计学意义(P>0.05)。结论:温补肾阳汤可能通过降低哮喘小鼠体内IL-5、SCF水平,抑制EOS分化募集,从而减轻EOS在肺组织局部的浸润。温补肾阳汤与醋酸泼尼松有一定的协同作用,联合使用可减少激素的用量。Objective:To explore the effects of wenbu shenyang (warming and tonifying kidney-yang) decoction on eosinophilic granulocyte (EOS) count of bone marrow suspension,serum IL-5 and stem cell factor(SCF) of lung tissue in asthma mice.Methods:50 mice were divided into five groups,normal,model,prednisone,wenbu shenyang decoction and prednisone plus wenbu shenyang decoction groups,each group contained 10 mice.Asthma models were made according to literature method,each group was given the corresponding medicine from the 29th day after modeling,once pre day for 14 days,than calculating EOS count of bone marrow suspension,observing the pathological changes of mice’s lung tissue by light microscope,serum IL-5 was determined by ELISA,SCF protein expression in lung tissue was determined by immunohistochemistry.Results:To compare with model group,EOS count of bone marrow suspension in prednisone,wenbu shenyang decoction and prednisone plus wenbu shenyang decoction groups was obviously decrease,serum IL-5 content and SCF expression in lung tissue were significantly reduced.To compare with wenbu shenyang decoction group,EOS count of bone marrow suspension in prednisone and prednisone plus wenbu shenyang decoction groups was obviously decrease,serum IL-5 content and SCF expression in lung tissue were significantly reduced.There was no obvious difference between prednisone and prednisone plus wenbu shenyang decoction groups on these observed indexes.Conclusion:Wenbu shenyang decoction can lower IL-5 and SCF levels of asthma mice,inhibit EOS differentiation,there was synergy effect between wenbu shenyang decoction and prednisone,the two combining application can reduce the dose of glucocorticosteroid.
