摘  要:目的观察激光诱导小鼠脉络膜新生血管(choroidal neovascularization,CNV)中膜攻击物与血管生长因子的表达。方法雄性C57BL/6小鼠66只,随机分为对照组与眼镜蛇毒因子(cobra venom factor,CVF)预处理组,每组各33只。用氪红激光诱导小鼠CNV动物模型;免疫组织化学检测激光诱导后24h视网膜色素上皮-脉络膜-巩膜复合体补体膜攻击复合物(membrane attack complex,MAC)的表达;RT-PCR分析激光诱导后1d、3d、5d、7d血管内皮生长因子(vascular endothelial growth factor,VEGF)mRNA表达;Western blot检测激光诱导后3dVEGF蛋白的表达。结果 CVF预处理组:激光诱导后24h,未见MAC阳性染色;激光诱导后3d、5d、7d与激光诱导后1d相比未见VEGF mRNA表达增加;激光7d未见CNV形成。对照组:激光诱导后24hMAC呈阳性染色;激光诱导后3d、5d、7d与激光诱导后1d相比VEGFmRNA分别增加380%、276%、98%(均为P<0.01);激光后3d,对照组较CVF预处理VEGF蛋白高表达。结论 CNV形成与补体MAC形成、VEGF表达上调相关联;MAC沉积导致VEGF表达增加;VEGF表达上调是CNV形成的主要原因。Objective To observe the expressions of membrance attack complex and vascular endothelial growth factor in Laser-Induced choroidal neovasculature (CNV) in mice. Methods A total of 66 male C57BL/6 mice were randomly and evenly divided into control group and cobra venom factor(CVF) pretreatment group. CNV was induced by laser photocoagulation with the krypton red laser. The expression of RPE- choroid-scleral membrance attack complex(MAC) at postoperative 24 hours was detec- ted by immunohistochemistry. The expression of vascular endothelial growth factor (VEGF) mRNA at postoperative 1 day,3 days,and 5 days was examined by RT-PCR. The expression of VEGF protein at postoperative 3 days was detected by Western blot. Results CVF pretreatment group:no staining of MAC was observed at postoperative 24 hours;compared with the postoperstive 1 day, the expressions of VEGF mRNA did not increace at postoperative 1 day,3 days, and 5 days;CNV did not form at postopera- tive 7 days. Control group:the strong staining of MAC was observed at postoperative 1 day; compared with the postoperstive 1 day, the expressions of VEGF mRNA at postop- erative 1 day, 3 days, and 5 days increaced 380% , 275% , and 98% respectively ( all P 〈 0. 01 ) ;There was high expression of VEGF protein at postoperative 3 days;CNV was in- duced at postoperative 7 days. Conelusion The formation of CNV is positively corre- lated to the formation of complement MAC and the up-regulaion expression of VEGF;the deposition of MAC leads to the increasing expression of VEGF;the increasing expression of VEGF is the main reason for the formation of CNV.
