摘 要:目的:研究雷公藤内酯醇(triptolide,TPL)对人淋巴瘤Raji细胞增殖抑制和诱导凋亡的作用。方法:体外培养人淋巴瘤Raji细胞,设不同浓度TPL作用不同时间处理Raji细胞的实验组和对照组。采用CCK-8法检测细胞增殖抑制率。经瑞氏-姬母萨染色,光学显微镜下观察雷公藤内酯醇处理后细胞的形态变化。应用流式细胞仪检测细胞的凋亡率和细胞周期分布。结果:CCK-8法实验表明,实验组TPL各浓度作用于Raji细胞24、48及72h后抑制率为11.02%-81.05%,呈时间、剂量依赖。细胞形态学检查表明,TPL各浓度作用于Raji细胞24h后细胞呈现典型的凋亡细胞的形态特征:光镜下可见细胞胞体固缩、核固缩、核碎裂及凋亡小体。流式细胞术检测显示,TPL各浓度作用于Raji细胞24h后出现明显的凋亡峰。细胞周期分析显示,TPL各浓度作用于Raji细胞24h后,TPL可阻滞Raji细胞于G0/G1期。结论:TPL对Raji细胞有增殖抑制和诱导细胞凋亡的作用,且呈时间、剂量依赖。其机制可能与细胞周期阻滞有关。Objective:To investigate the effects of triptolide(TPL) on the proliferation and apoptosis of human lymphoma cell line Raji.Methods:Human lymphoma cell line Raji was cultivated in vitro.Two groups were set up as the control group and the experiment group that was treated with various concentrations and time of TPL.The growth of the cells was tested by CCK-8 assay.The morphologic changes of Raji cells induced by TPL were observed under light microscopy with Wright-Giemsa staining.Cell cycle and cell apoptosis were analyzed by cytometry(FCM).Results:CCK-8 showed that in the experimental group,the inhibitive rates of Raji after treated with various concentrations of TPL for 24,48,72 hours were 11.02%-81.05%.The Raji cells were inhibited in both dose-dependent and time-dependent.After treated with different concentrations TPL for 24h,typical apoptotic morphologic features appeared in Raji cells of experimental group:condensation of cells and nuclear chromatin,disintegration of nuclear chromatin,and apoptotic bodies.FCM showed that apoptosis peak emerged significantly after Raji cells treated with TPL for 24 hours in the experimental group.After Raji cells treated with various concentrations of TPL for 24 hours in the experiment group,the cell percentage in G0/G1 phase of the cell cycle distribution existed a significant difference compared with the control group.Conclusion:TPL could inhibit the proliferation and induce apoptosis of Raji cells in a time-dose dependent manner.It may be related to cell cycle arresting.
