摘 要:目的研究氟化钠对大鼠睾丸生精细胞Caspase-3和Caspase-9表达的影响,探讨其在生精细胞凋亡调控中的作用。方法雄性Wistar大鼠,随机分为4组,对照组、低氟组、中氟组和高氟组,对照组饮去离子水,低氟组、中氟组和高氟组饮用分别含50、100和200 mg/L氟化钠去离子水。染毒120 d后处死动物。采用免疫组化法(SP法),观察不同剂量染毒后睾丸生精细胞中Caspase-3和Caspase-9的表达情况。结果所有染氟组均观察到生精细胞凋亡和Caspase-3及Caspase-9表达的变化,50、100和200 mg/L染毒120 d后,大鼠睾丸生精细胞中Caspase-3和Caspase-9的表达水平不同程度的上升,尤其以200 mg/L组最为明显。结论本试验条件下,氟化钠可能通过影响大鼠睾丸生精细胞中Caspase-3和Caspase-9的释放,调控生精细胞凋亡。Objective To investigate the effects of sodium fluoride on the expressions of Caspase-3 and Caspase-9 in spermatogenic cells of rat testis,and to explore the mechanism of apoptosis regulation in spermatogenic cells. Methods The rats exposed to sodium fluoride were treated with 50, 100, 200 mg/L doses in water for 120 days,the control group drunk deionized water. Twelve mice in each group were put to death at 120 days and then testes were separated from rats. The Caspase-3 and Caspase-9 protein expressions in the spermatogenic cells of rat testis were examined by immunohistochemical technique ( SP). Results In comparison with the control group, the expressions of Caspase-3 and Caspase-9 proteins in spermatogenic cells increased, especially in the group of 200 mg/L dose. Conclusion It was suggested that sodium fluoride could induce the apoptosis of spermatogenic cells by accelerating the release of Caspase-3 and Caspase-9.
