染色剂浓度对复方白及乳膏中多糖定量的影响

摘　　要：目的考察0.1%～0.4%蒽酮-硫酸试液对复方白及乳膏中多糖含量测定结果的影响,筛选最佳染色剂浓度与用量,提高检出效率。方法用无水葡萄糖为对照品,0.1%～0.4%蒽酮-硫酸试液为染色剂分组染色复方白及乳膏供试液,分光光度法测定含量,测定波长为620 nm,比较各组吸光度、含量测定结果、回收率。结果在4组染色剂中,0.2%蒽酮-硫酸试液用量在6 mL时吸光度、含量测定、回收率均优于其他组,其中回收率为96.0%,RSD=1.22%(n=6),检出效果依次为:0.2%组>0.1%组>0.3%组>0.4%组。结论染色剂浓度对测定结果有较大影响,使用0.2%浓度的染色剂检出率高,重现性好,适用于复方白及乳膏中多糖的含量测定。Objective To investigate the influence of 0.1%~0.4% of anthrone-sulfuric on the polysaccharide content determination of compound of Bletilla Striata cream,and to select the optimal concentration of dye and the dosage,then to improve the detective efficiency.Methods Anhydrous dextrose was used as a reference substance,0.1%~0.4% of anthrone-sulfuric was used as stain to dye compond Bletilla Striata cream,and the content was determined by spectrophotometry.The detective wavelength was set at 620 nm.Absorbance,concentration and recovery rate of each group were compared.Results The content determination results and recovery were better in the group with 6 mL of 0.2% anthrone-sulfuric,and the recovery rate was 96.0% with RSD of 1.22（n=6）.The detective effect strength was ordered by the concentration of anthrone-sulfuric：0.2%0.1%0.3%0.4%.Conclusion The dye concentration can influence the content determination,the detective rate was highest when the dye concentration was 0.2% with good reproducibility,and it can be applied to determine the content of compound of Bletilla Striata cream.
