白细胞介素-1受体相关激酶-4在血吸虫病门脉高压小鼠肠系膜静脉的表达变化及其意义

摘　　要：目的探讨血吸虫病门脉高压小鼠肠系膜静脉组织白细胞介素-1受体相关激酶-4（IRAK-4）的表达变化及意义。方法将55只雄性昆明小鼠随机分为对照组（n=15）和感染组（n=40）。感染组采用腹部敷贴法感染日本血吸虫尾蚴，分别于感染后12、20周随机分为12周组（n=20）、20周组（n=20）；采用逆转录-聚合酶链反应（RT—PCR）法检测肠系膜静脉IRAK-4mRNA表达与肿瘤坏死因子（TNF）-α mRNA表达，Westernblot法检测肠系膜静脉IRAK-4蛋白表达。结果感染组小鼠肠系膜静脉IRAK-4mRNA表达强度值（12周组0．734±0．081，20周组0．682±0．069）和IRAK-4蛋白表达水平（12周组：1034．52±89．72，20周组：989．56±69．35）较对照组（IRAK-4mRNA表达强度值0．163±0．031，IRAK4蛋白表达水平667．45±72．22）明显升高（P〈0．01，P〈0．05）；20周组IRAK-4mRNA和蛋白表达与12周组比较差异无统计学意义（P〉0．05）；各组IRAK-4mRNA的表达与其效应分子TNF-αmRNA的表达呈显著正相关（r=5．56，P〈0．01）。结论门静脉高压症小鼠内脏血管IRAK-4在转录和翻译水平的表达升高，并激活下游炎性反应，是参与门静脉高压性血管病变的重要因素。Objective：To explore the expression and significance of interleukin-1 receptor associ- ated kinase-4 （IRAK-4） in mesenteric vein of rats with schistosomal cirrhosis. Methods Fifty-five rats were randomly divided into two groups： normal control group （N group, n = 15 ）） and model group （M group, n =40）. The rats in M group were infected percutaneously with cercariae of schistosomiasis japoni- ca, and divided into post-infection 12-week subgroup and 20-week subgroup. The mRNA expression levels of IRAK-4 and tumor necrosis factor （TNF）-ct were detected by using reverse transcriptase polymerase chain reaction （RT-PCR）. Western blotting was applied to examined the protein expression of IRAK-4. Results The expression of IRAK-4 mRNA in mesenteric vein in M groups （12th week： 0. 734 ± 0. 081; 20th week： 0. 682 ±0. 069） was significantly stronger than in N group （0. 163 ±0. 031 ）. The expression of IRAK-4 protein in M group （ 12th week ： 1034. 52 ± 89.72 ; 20th week ： 989. 56 ± 69. 35 ） was signifi- candy stronger than in N group （667.45 ± 72. 22）. No significant differences were found in the level of IRAK-4 between post-infection 20-week group and post-infection 12-week group （ P 〉 0. 05 ）. The expres- sion of IRAK-4 mRNA was significantly positively correlated with the level of TNF-α mRNA （ r = 5.56 ,P 〈 0. 01 ）. Conclusion The high expression of IRAK-4 and activation of downstream inflammatory factor play an important role in vasculopahty of rats with schistosomal cirrhosis.
